基于16S rRNA测序探究黄芪润肠汤对大肠黑变病肠道菌群的调控作用OA
Astragalus Decoction for moistening intestines treats melanosis coli via regulating intestinal flora based on 16S rRNA sequencing
目的 观察黄芪润肠汤对大黄所致大肠黑变病大鼠肠黏膜损伤及肠道微生物的影响,探讨黄芪润肠汤治疗大肠黑变病的作用机制.方法 将 60 只 SD 大鼠随机分为空白组、模型组、黄芪润肠汤低剂量组、黄芪润肠汤高剂量组以及乳果糖组,每组12 只.除空白组外,其余组采用生大黄液连续灌胃30 d 方法建立大肠黑变病模型.确认造模成功后,黄芪润肠汤低、高剂量组分别按照0.75 mL/100 g 和1.5 mL/100 g 的剂量给予黄芪润肠汤灌胃,乳果糖组按照1 g/kg 剂量给予乳果糖口服溶液灌胃,均1 次/d,连续灌胃 28 d.观察各组大鼠结肠组织大体情况,HE 染色观察结肠组织病理形态,采集各组粪便进行 16S rRNA 测序分析.结果 模型组大鼠结肠组织出现明显的色素沉淀;结肠黏膜上皮细胞大量脱落,固有层及黏膜下层明显可见褐色色素沉积;与模型组比较,各药物组大鼠结肠组织黑色素沉淀减少,结肠黏膜上皮细胞少量脱落,结肠黏膜固有层及黏膜下层褐色色素沉积减少,其中黄芪润肠汤高剂量组和乳果糖组改善更明显.粪便 Alpha 多样性分析显示,黄芪润肠汤高剂量组和乳果糖组结肠菌群丰度和多样性明显提高;Beta 多样性PCA 分析显示,模型组与其他组样本明显分开;物种分类显示,黄芪润肠汤低、高剂量组嗜黏蛋白阿克曼菌(Akkermansia muciniphila)、马西福卡埃科拉(Phocaeicola massiliensis)、普通拟杆菌(Pho-caeicola vulgatus)和类阿氏动丝菌(Kineothrix alysoides)的丰度均不同程度提高.结论 黄芪润肠汤能够调控肠道菌群来治疗大肠黑变病.
Objective It is to observe the effect of Astragalus Decoction for moistening intestines(ADMI)on intestinal mucosal injury and gut microbiota in rats with rhubarb-induced melanosis coli(MC),and to explore the mechanism of ac-tion of this drug in the treatment of MC.Methods Sixty SD rats were randomly divided into normal group,model group,low-dose and high-dose groups of ADMI,lactulose group,with 12 rats in each group.The rats of all groups except for the blank group were continuously fed with raw rhubarb solution via gastric administration for 30 days to establish models of MC.After successful modeling,the low-dose and high-dose groups of ADMI were administered ADMI via gastric gavage at the doses of 0.75 mL/100 g and1.5 mL/100 g,respectively,while the lactulose group was administered an oral lactulose solution at a dose of 1 g/kg,all once daily,continuously treated for 28 days.The gross appearance of colon tissue from rats in each group was examined,the histopathology of the colon tissue was observed by HE staining,and fecal samples were collected from each group for 16S rRNA sequencing analysis.Results Significant pigment deposition in colon tissue was ob-served in the rats of the model group;there was extensive shedding of colonic mucosal epithelial cells,and brown pigment deposits were clearly visible in the lamina propria and submucosa;compared with the model group,pigment deposition in colon tissue and brown pigment deposition in the lamina propria and submucosa of colon were reduced with minimal shed-ding of colonic mucosal epithelial cells in the rats of the medicated groups,and the improvements were more significant in the high-dose group of ADMI and lactulose group.Analysis of fecal alpha diversity showed that the colonic microbial abun-dance and diversity were significantly increased in the high-dose group of ADMI and lactulose group;PCA analysis of beta diversity revealed that the samples in the model group were clearly distinct from those in the other groups;species-level a-nalysis showed that the abundances of Akkermansia muciniphila,Phocaeicola massiliensis,Phocaeicola vulgatus,and Kineo-thrix alysoides were all increased to varying degrees in both the low-dose and high-dose groups of ADMI.Conclusion ADMI can treat MC via regulating intestinal flora.
