芪苈强心胶囊对兔心力衰竭模型电重构影响的研究OA
Effect of Qiliqiangxin Capsule on electrical remodeling in a rabbit heart failure model
目的 应用微电极阵列(MEA)技术研究芪苈强心胶囊对兔心力衰竭(心衰)模型电重构的影响.方法 选取新西兰兔60只,随机分成6组:对照组、心衰组、芪苈强心A组(10 mg/kg)、芪苈强心B组(20 mg/kg)、芪苈强心C组(40 mg/kg)、芪苈强心D组(80 mg/kg),各组均10只.心衰造模成功后,迅速开胸取出心脏,行langendroff灌流,用柔性电极贴附心室肌,分别记录各组场电位时程(fAPD)、兴奋传导速度(CV)和心率(HR)的变化.结果 与对照组比较,心衰组兔心室肌fAPD显著延长,CV减慢,HR明显加快,差异均有统计学意义(P<0.05).与心衰组比较,芪苈强心各组兔心室肌fAPD减慢,CV加快,HR减慢,差异均有统计学意义(P<0.05),且效应呈浓度依赖性增强.结论 芪苈强心胶囊可明显改善兔心衰模型电重构.
Objective To investigate the effect of Qiliqiangxin Capsule on electrical remodeling in a rab-bit heart failure model using microelectrode array(MEA)technology.Methods Sixty New Zealand rabbits were randomly divided into 6 groups(n=10):control group,heart failure group,Qiliqiangxin A group(10 mg/kg),Qiliqiangxin B group(20 mg/kg),Qiliqiangxin C group(40 mg/kg),and Qiliqiangxin D group(80 mg/kg).After successful heart failure modeling,the hearts were rapidly excised and subjected to langendorff perfusion.Flexible electrodes were attached to the ventricular myocardium to record changes in field action po-tential duration(fAPD),conduction velocity(CV),and heart rate(HR)in each group.Results Compared with the control group,the fAPD in the ventricular myocardium of the heart failure group was significantly pro-longed,CV was slowed,and HR was significantly accelerated,with statistically significant differences(P<0.05).Compared with the heart failure group,fAPD of ventricular myocardium was shortened,CV was accel-erated,and HR was slowed in all Qiliqiangxin groups,the differences were all statistically significant(P<0.05),with the effect showing a concentration-dependent enhancement.Conclusion Qiliqiangxin Capsule can significantly improve electrical remodeling in a rabbit heart failure model.
