L-异亮氨酸生产菌的苏氨酸脱水酶特性及应用OA

苏氨酸脱水酶(threonine dehydratase,TD)是α-酮丁酸和 L-异亮氨酸生物合成途径中的关键酶,其活性受 L-异亮氨酸的反馈抑制.对来源于 L-异亮氨酸生产菌株谷氨酸棒杆菌(Corynebacterium glutamicum)YI 的苏氨酸脱水酶TDYI进行序列分析,发现其 363 位 L-脯氨酸突变为 L-亮氨酸(P363L).酶学实验结果表明,该酶解除了 L-异亮氨酸的反馈抑制作用.TDYI 的米氏常数(Km)、最大反应速率(vmax)、催化常数(Kcat)及 Kcat/Km 分别为 8.7 mmol/L、88.2 μmol/(min·mg)、9.2 s-1 和 1.06 L/(mmol·s),其最适 pH 和温度分别为 7.0 和 35℃,与野生型 TD 无明显差异,表明P363L 突变对 TDYI的催化性能无显著影响.将 TDYI应用于α-酮丁酸和 L-异亮氨酸的合成,其产量分别达到 13.3 g/L和 5.3 g/L,且 TDYI效果优于大肠杆菌来源的抗 L-异亮氨酸反馈抑制的 TDM.本研究从 L-异亮氨酸生产菌株 C.glu-tamicum YI中挖掘出解除L-异亮氨酸反馈抑制的TDYI,可为α-酮丁酸、L-异亮氨酸及其衍生物的生物合成提供参考.

Threonine dehydratase(TD)is a key enzyme in the biosynthesis pathways of α-ketobutyrate and L-isoleucine,and its activity is subject to the feedback inhibition by L-isoleucine.In this study,sequence of TDYI from the L-isoleucine produc-ing strain Corynebacterium glutamicum YI was analyzed.The L-proline at 363rd site was substituted by L-leucine(P363L)in TDYI.Enzymatic assays demonstrated that the feedback inhibition by L-isoleucine of TDYI was abolished.Meanwhile,TDYI exhibited a Km of 8.7 mmol/L,a vmax of 88.2 μmol/(min·mg),a Kcat of 9.2 s-1,and a Kcat/Km of 1.06 L/(mmol·s).Its optimal pH and temperature were 7.0 and 35℃,respectively,similar with those of the wild-type TD,indicating that the P363L muta-tion did not markedly affect the catalytic performance of TDYI.Subsequently,TDYI were applied in biosynthesis of α-ketobutyrate and L-isoleucine,achieving a production of 13.3 and 5.3 g/L,respectively,significantly higher than those of the strains harboring TDM derived from Escherichia coli.This study identified and characterized TDYI,a feedback-inhibition-relieved threonine dehydratase from the industrial strain C.glutamicum YI.The findings provide a valuable reference for the biosynthesis of α-ketobutyrate,L-isoleucine,and their related products.
