TGF-β2通过增加线粒体活性氧积累和阻滞细胞周期促进猪间充质干细胞衰老OA
TGF-β2-Induced Senescence of Porcine Bone Marrow Mesenchymal Stem Cells via Mitochondrial Reactive Oxygen Species Accumulation and Cell Cycle Arrest
本研究旨在探讨转化生长因子-β2(TGF-β2)在猪骨髓间充质干细胞(pBMMSCs)衰老中的作用及机制.首先分析 pBMMSCs 在体外复制性衰老过程中 TGF-β2 水平的变化,研究 1 ng/mL、10 ng/mL 和 100 ng/mL 的 TGF-β2 对细胞生长、增殖、衰老、线粒体活性氧(mtROS)水平、细胞周期及相关基因表达的影响.结果显示,pBMMSCs 表达干细胞标志物 CD90 和 CD44,具备成脂和成骨分化潜能.随着传代次数的增加,TGF-β2 的 mRNA 水平显著升高,蛋白质的质量浓度从(41.97±9.46)pg/mL 上升到(77.04±3.44)pg/mL.TGF-β2 处理后,细胞增殖能力下降,衰老细胞显著增加,mtROS 水平升高,细胞周期阻滞在 G1 期,OCT4、Ki67、PCNA、SOD2 表达下降,P53 和 P21 表达上升.上述结果表明,TGF-β2 通过促进线粒体活性氧积累并阻滞细胞周期 G1 期,诱导 pBMMSCs 衰老.本研究为深入理解 TGF-β2 在间充质干细胞衰老过程中的作用提供了参考.
This study aimed to investigate the role and mechanism of transforming growth factor-beta 2(TGF-β2)in the senescence of porcine bone marrow mesenchymal stem cells(pBMMSCs).First,changes in TGF-β2 levels during in vitro replicative senescence of pBMMSCs were analyzed.Then,the effects of TGF-β2 at concentrations of 1 ng/mL,10 ng/mL,and 100 ng/mL on cell growth,proliferation,senescence,mitochondrial reactive oxygen species(mtROS)levels,cell cycle,and related gene expression were also examined.The results showed that pBMMSCs expressed stem cell markers CD90 and CD44,and had the potential for adipogenic and osteogenic differentiation.With increasing passage numbers,the mRNA levels of TGF-β2 significantly increased,and its protein content rose from(41.97±9.46)pg/mL to(77.04±3.44)pg/mL.After TGF-β2 treatment,cell proliferation ability decreased,the number of senescent cells significantly increased,mtROS levels were elevated,and the cell cycle was arrested at G1 phase.Moreover,the expression of OCT4,Ki67,PCNA,and SOD2 was downregulated,while the expression of P53 and P21 was upregulated.These findings indicated that TGF-β2 in-duced the aging of pBMMSCs by promoting mtROS accumulation and causing G1-phase cell cycle arrest.This study pro-vides new insights into the role of TGF-β2 in the senescence process of mesenchymal stem cells.
