IQCN在人睾丸和精子中的表达及其与精子活力的相关性分析OA
Expression of IQCN in human testes and sperm and its correlation with sperm motility
目的 探讨IQCN(IQ motif containing N)在人类睾丸和精子中的表达情况及其与精子活力的相关性.方法 检索哺乳动物生殖遗传数据库,了解IQCN在睾丸组织和生殖细胞的表达情况.通过Western blotting分析IQCN蛋白在人睾丸和精子上的表达水平及IQCN在不同精子活力组间的蛋白表达差异,采用免疫组化、免疫荧光实验检测IQCN在人睾丸和精子中的表达定位情况,并通过生物信息学分析IQCN可能的生物学功能和相关作用机制.结果 IQCN在人睾丸组织及精子中均有表达,且特异性表达于精子顶体位置.与正常活力组相比,弱精子症组精子的IQCN表达水平显著下降(P<0.05).生物信息学分析发现IQCN可以与CALM2、CALM3、CALML3、PICALM等钙调蛋白基因相互作用,通过参与Ca2+信号通路、PKC信号通路、精子鞭毛形成组装等生物学过程来发挥作用.结论 IQCN在人睾丸和精子中有表达,且在不同精子活力组间表达具有显著差异,IQCN表达异常可能导致人类精子活力下降和男性不育,值得进一步深入研究.
Objectives:To investigate the expression of the IQCN(IQ motif containing N)gene in human testicular tissue and spermatozoa,and its correlation with human sperm motility. Methods:The mammalian reproductive genetics database was searched to examine the expression of IQCN in testicular tissue and germ cells.Western blotting was used to detect the expression levels of IQCN protein in human testes and sperm,and the protein expression differences of IQCN among different sperm motility groups.Immunohistochemistry and immunofluorescence were employed to study the localization of IQCN expression in human testis and sperm.Bioinformatic analysis was conducted to explore the potential biological functions and related mechanisms of IQCN. Results:IQCN was expressed in both human testicular tissue and sperm,specifically localized in the sperm acrosome.Compared with the normal motility group,the expression level of IQCN was significantly reduced in the asthenozoospermia group.Bioinformatic analysis revealed that IQCN could interact with calmodulin genes such as CALM2,CALM3,CALML3,and PICALM,and may play a role through involvement in biological processes including Ca2+signaling pathways,PKC signaling pathways,and sperm flagellum formation and assembly. Conclusions:IQCN is expressed in human testis and sperm,and its expression differs significantly among groups with varying sperm motility.Abnormal expression of IQCN may lead to decreased sperm motility and male infertility,warranting further in-depth investigation.
