基于UPLC-Q Exactive Plus Orbitrap MS和UPLC-MS的益脑膏化学成分定性和定量研究OA
Qualitative and quantitative study of chemical constituents of Yinao Ointment(益脑膏)based on UPLC-Q Exactive Plus Orbitrap MS and UPLC-MS
目的 建立益脑膏化学成分的定性与定量分析方法,明确其物质基础.方法 采用Thermo Fisher Scientfic Accucore Phenyl Hexyl色谱柱(2.1 mm×100 mm,2.6 μm);以0.1%甲酸水溶液-乙腈为流动相进行梯度洗脱;流速0.2 mL·min-1;柱温35℃;采用加热型电喷雾离子源,采用全扫描/数据依赖二级扫描(Full Scan/ddMS²),结合对照品保留时间、质谱离子信息以及数据库与文献中的相关质谱信息进行比对,识别益脑膏中的化学成分;进一步采用UPLC-MS技术,结合多反应监测模式(MRM)扫描,建立能同时测定益脑膏中24个成分含量的方法.结果 通过对照品比对及高分辨质谱解析,鉴定出116个成分并解析了其裂解规律.在此基础上,建立了能同时测定益脑膏中24个成分的UPLC-MS定量方法,上述的24个成分在各自检测质量浓度范围内线性关系良好(r>0.9992),精密度、重复性及稳定性的RSD均小于6.46%,平均加样回收率96%-103.90%(RSD均≤5.50%).结论 所建立的定性方法能快速、灵敏、准确的鉴定益脑膏中的化学成分,所建立的定量方法灵敏可靠,专属性强,能全面反映益脑膏中各类成分的含量,可为益脑膏的品质评价与质量提升提供参考依据.
Objective To establish qualitative and quantitative analysis methods for the chemical constituents of Yinao Ointment(益脑膏,YNO)and clarify its material basis.Methods Chromatographic separation was performed on a Thermo Fisher Scientific Accucore Phenyl Hexyl column(2.1 mm×100 mm,2.6 μm)with gradient elution using 0.1%formic acid aqueous solution-acetonitrile as the mobile phase at a flow rate of 0.2 mL/min.The column temperature was maintained at 35 ℃.Using a heated electrospray ionization source,chemical constituents in YNO were identified through Full Scan/ddMS² mode,combined with comparisons of retention times of reference substances,mass spectrometry ion information,and relevant mass spectrometry data from databases and literature.Further-more,an UPLC-MS method was developed for the simultaneous determination of 24 components in YNO using multiple reaction monitor-ing(MRM)mode scanning.Results Through comparison with reference substances and high-resolution mass spectrometry analysis,116 components were identified and their fragmentation patterns were elucidated.On this basis,an UPLC-MS quantitative method was established for the simultaneous determination of 24 components in YNO.These 24 components showed good linear relationships within their respective detection concentration ranges(r>0.9992).The RSDs for precision,repeatability,and stability were all less than 6.46%,and the average recoveries ranged from 96%to 103.90%(all RSDs≤5.50%).Conclusion The established qualitative method can rapidly,sensitively,and accurately identify the chemical constituents in YNO.The established quantitative method is sensitive,reliable,and specific,capable of comprehensively reflecting the contents of various components in YNO,thus providing a reference basis for quality evaluation and improvement of YNO.
