清半夏对OVA小鼠的抗炎祛痰作用及机制研究OA
Study on the anti-inflammatory and expectorant effects and mechanism of Pinelliae Rhizoma Praeparatum Cum Alumine on OVA mice
目的 探究半夏炮制品清半夏(PRA)的水提物(PRAWE)对卵清蛋白(OVA)刺激哮喘小鼠的抗炎及祛痰作用及机制.方法 HE、Masson、AB-PAS染色切片观察小鼠肺组织病理学变化;应用非靶向代谢组学技术分析PRAWE对OVA刺激小鼠的代谢调节;ELISA试剂盒检测小鼠血清中免疫球蛋白E(IgE),肺泡灌洗液中肿瘤坏死因子-α(TNF-α)、白介素(IL)-6、IL-1β含量;Western blot法对小鼠肺组织中核因子κB(NF-κB)/核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)信号通路相关蛋白进行检测.结果 与OVA组小鼠比较,PRAWE各剂量组小鼠肺部组织中炎症浸润情况,胶原纤维沉积现象减轻,粘液分泌量下调.通过影响核苷酸、嘧啶代谢等途径调节OVA小鼠肺组织代谢紊乱;ELISA试剂盒结果显示,与OVA组小鼠比较,PRAWE下调了OVA小鼠血清中IgE,肺泡灌洗液中TNF-α、IL-6、IL-1β含量(P<0.01,P<0.05);Western blot结果显示,PRAWE显著抑制磷酸化(p)p65、p-NF-κB抑制因子α(IκBα)和NLRP3、凋亡相关斑点样蛋白(ASC)、半胱氨酸天冬氨酸蛋白酶-1(Caspase-1)的表达(P<0.05,P<0.01).结论 PRAWE能有效缓解OVA刺激小鼠体内炎症,减少粘液分泌量,调节小鼠体内代谢紊乱,通过影响NF-κB/NLRP3信号通路发挥对OVA哮喘小鼠的抗炎及祛痰作用.
Objective To investigate the anti-inflammatory and expectorant effects and mechanism of the water extract of Pinelliae Rhi-zoma Praeparatum Cum Alumine(PRAWE),a processed product of Rhizoma Pinelliae,in ovalbumin(OVA)-induced asthmatic mice.Methods HE staining,Masson staining,and AB-PAS staining were used to observe the pathological changes in mouse lung tissues.Untargeted metabolomics technology was applied to analyze the metabolic regulation of PRAWE in OVA-induced mice.ELISA kits were used to detect the levels of immunoglobulin E(IgE)in serum,and tumor necrosis factor-α(TNF-α),interleukin(IL)-6,and IL-1β in bronchoalveolar lavage fluid.Western blot(WB)was performed to detect the expression of proteins related to the nuclear factor kappa B(NF-κB)/NOD-like receptor protein 3(NLRP3)signaling pathway in mouse lung tissues.Results Compared with the OVA group,PRAWE treatment at various doses alleviated inflammatory infiltration,reduced collagen fiber deposition,and decreased mucus secre-tion in the lung tissues of mice.PRAWE regulated metabolic disorders in the lung tissues of OVA-induced mice by affecting pathways such as nucleotide metabolism and pyrimidine metabolism.ELISA results showed that compared with the OVA group,PRAWE down-regulated the levels of IgE in serum and TNF-α,IL-6,and IL-1β in bronchoalveolar lavage fluid(P<0.01 or P<0.05).WB results showed that PRAWE significantly inhibited the expression of phosphorylated(p)-p65,p-inhibitor of kappa B alpha(IκBα),NLRP3,apoptosis-associated speck-like protein containing a CARD(ASC),and caspase-1(P<0.05 or P<0.01).Conclusion PRAWE can effectively alleviate inflammation,reduce mucus secretion,and regulate metabolic disorders in OVA-induced mice.It exerts anti-inflammatory and expectorant effects in OVA-induced asthmatic mice by influencing the NF-κB/NLRP3 signaling pathway.
