目的 研究智元康颗粒(ZYK)对APP/PS1小鼠神经保护作用,并探讨其调节分选连接蛋白A(SorLA)抑制淀粉样前体蛋白(APP)异常水解的作用机制.方法 采用Morris水迷宫和新物体识别检测学习记忆功能,刚果红染色检测β-淀粉样蛋白沉积,甘氨酸镀银检测缠结,Western blot、RT-qPCR 检测SorLA通路及其下游分子的表达情况.结果 与Control组比较,Model组小鼠学习记忆能力明显下降,神经损伤严重(P<0.01);海马SorLA、ADAM10、GGA3 mRNA和蛋白表达水平下降,BACE1、TNF-α、Iba1 mRNA和蛋白表达水平上升(P<0.01),PSD95、SYN蛋白表达水平下降,APP表达水平上升(P<0.01).与Model组比较,ZYK组与盐酸多奈哌齐(Donepezil)组学习认知功能明显改善;刚果红染色、甘氨酸镀银染色结果显示Aβ和Tau病理、神经损伤程度明显改善(P<0.05,P<0.01);RT-qPCR和Western blot显示ZYK组SorLA、ADAM10、GGA3 mRNA和蛋白表达水平上升,BACE1、TNF-α、Iba1 mRNA和蛋白表达水平下降,PSD95、SYN蛋白表达水平上升,APP蛋白表达水平下降(P<0.05,P<0.01);Donepezil组PSD95、SYN、GGA3表达水平上升、APP、BACE1、TNF-α、Iba1表达水平下降(P<0.05,P<0.01),SorLA、ADAM10表达水平无统计学意义(P>0.05).结论 智元康颗粒可通过调节SorLA来抑制APP的异常水解减轻神经炎症、发挥神经保护作用.

Objective To investigate the neuroprotective effect of Zhiyuankang Granule(智 元 康 颗 粒,ZYK)on APP/PS1 mice and explore its mechanism of action in regulating sorting protein-related receptor with A-type repeats(SorLA)to inhibit abnormal hydrolysis of amyloid precursor protein(APP).Methods Learning and memory functions were assessed using the Morris water maze and novel object recognition tests.β-amyloid deposition was detected by Congo red staining,and tangles were examined by glycine silver staining.The expression levels of the SorLA pathway and its downstream molecules were measured via Western blot(WB)and RT-qPCR.Results Compared with the Control group,the Model group showed significant decline in learning and memory abilities and severe neural damage(P<0.01).The mRNA and protein expression levels of SorLA,ADAM10,and GGA3 in the hippocampus were decreased,while those of BACE1,TNF-α,and Iba1 were increased(P<0.01).The protein expression levels of PSD95 and SYN were decreased,and APP expression was increased(P<0.01).Compared with the Model group,both the ZYK group and the Donepezil hydrochloride group exhibited significantly improved learning and cognitive function.Congo red staining and glycine silver staining results indicated notable alleviation of Aβ and Tau pathology as well as neural damage(P<0.05,P<0.01).RT-qPCR and WB analysis revealed that the ZYK group had increased mRNA and protein expression levels of SorLA,ADAM10,and GGA3,decreased levels of BACE1,TNF-α,and Iba1,elevated protein expression of PSD95 and SYN,and reduced APP protein expression(P<0.05,P<0.01).In the Done-pezil group,the expression levels of PSD95,SYN,and GGA3 were increased,while those of APP,BACE1,TNF-α,and Iba1 were decreased(P<0.05,P<0.01);however,the expression levels of SorLA and ADAM10 showed no statistical significance(P>0.05).Conclusion ZYK can inhibit abnormal APP hydrolysis by regulating SorLA,thereby mitigating neuroinflammation and exerting a neuro-protective effect.
