基于标准汤剂的吴茱萸配方颗粒制备工艺研究OA
Study on preparation technology of Fructus Euodiae Formula Granule(吴茱萸配方)based on standard decoction
目的 以吴茱萸标准汤剂为基准,研究吴茱萸配方颗粒的制备工艺.方法 制备吴茱萸标准汤剂后,以出膏率和柠檬苦素、吴茱萸碱、吴茱萸次碱转移率为评价指标,以加水量、提取时间、提取次数为影响因素,正交试验优选提取工艺,考察浓缩温度、干燥方式、辅料量,优化配方颗粒制备工艺.结果 最佳提取工艺为8倍量水提取2次,每次1.5h;最佳浓缩工艺为70℃及以下减压浓缩,不超过9h;最佳干燥工艺为浓缩液密度1.03~1.05,喷雾干燥.添加0%~7%辅料,干法制粒.出膏率、各指标含量均与标准汤剂基本一致.结论 该实验以标准汤剂为指标对吴茱萸配方颗粒制备的生产工艺进行优化,保证了配方颗粒与吴茱萸传统汤剂质量的一致性,保障了临床用药的安全有效稳定可控,为吴茱萸以及其他中药配方颗粒的制备提供新思路.
Objective To study the preparation technology of Fructus Euodiae Formula Granule(吴茱萸配方,FEFG)based on its standard decoction.Methods After preparing Fructus Euodiae(FE)standard decoction,using extraction rate,transfer rates of limonin,evodiamine,and rutaecarpine as evaluation indicators,and taking water volume,extraction time,and extraction cycles as influencing factors,orthogonal experiments were conducted to optimize the extraction process and investigate concentration temperature,drying method,and excipient ratio to refine the formula granule preparation.Results The optimal extraction process involves using 8 times the amount of water for two extractions,each lasting 1.5 h.The optimal concentration process is vacuum concentration at 70℃ or below for no more than 9 h.The optimal drying process requires a concentrated liquid density of 1.03-1.05 g/cm³ followed by spray drying.With the addition of 0%-7% excipients,dry granulation is performed.Both the extract yield and content of various components are essentially consistent with those of the standard decoction.Conclusion This study optimized the production process of FEFG using standard decoc-tion as the quality benchmark,thereby ensuring the quality consistency between the formula granules and traditional FE decoction.The optimized process guarantees the safety,efficacy,stability,and controllability of clinical medication,while providing novel technical approaches for the preparation of FE and other Chinese herbal formula granules.
