参芪地黄汤化裁方含药血清对高糖诱导的小鼠永生化足细胞焦亡的保护作用影响OA
The protective effect of the serum containing the modified prescription of Shenqi Dihuang Decoction(参芪地黄汤)on high-glucose-induced pyroptosis in MPC5 cells
目的 观察参芪地黄汤化裁方含药血清对高糖诱导的小鼠永生化足细胞(MPC5)焦亡的保护作用.方法 体外培养小鼠足细胞,筛选MPC5细胞高糖损伤干预浓度与时间及参芪地黄汤化裁方含药血清的最佳干预浓度;将细胞分为正常对照组、高糖模型组、中药含药血清组、NLRP3抑制剂对照组(MCC950组)进行实验.光镜观察各组细胞焦亡形态差异;酶联免疫吸附测定法(ELISA)检测各组细胞上清IL-1β、IL-18水平;蛋白免疫印迹法(Western blot)检测细胞Nephrin、Podocin、PODXL、WT-1、Desmin、NLRP3、Cleaved Caspase-1、GSDMD-N蛋白表达水平;免疫荧光法检测Nephrin、NLRP3、GSDMD-N、Cleaved Caspase-1蛋白表达水平.结果 与正常对照组比较,高糖模型组部分细胞膜完整性破坏,细胞密度降低;Elisa结果显示IL-1β、IL-18表达水平上调;WB结果显示Nephrin、Podocin、PODXL、WT-1蛋白表达减少(P<0.05),Desmin、NLRP3、Cleaved Caspase-1、GSDMD-N表达增加(P<0.05);免疫荧光结果显示NLRP3、Cleaved Caspase-1、GSDMD-N相对荧光表达量增多(P<0.05),Nephrin相对荧光表达水平降低(P<0.05).中药含药血清组与NLRP3抑制剂对照组(MCC950组)上述情况较模型组改善(P<0.05).结论 参芪地黄汤化裁方含药血清可减轻高糖诱导的足细胞损伤,其机制与改善细胞焦亡有关.
Objective To investigate the protective effect of serum containing Shenqi Dihuang Decoction modified prescription(参芪地黄汤,SQDH)against pyroptosis in immortalized mouse podocytes induced by high glucose.Methods Mouse podocytes(MPC5)were cultured in vitro.Optimal intervention concentrations of high glucose and SQDH-containing serum,as well as exposure duration,were determined.Cells were divided into four groups:normal control,high glucose model,SQDH-containing serum,and NLRP3 inhibitor control(MCC950 group).Morphological changes indicative of pyroptosis were observed under light microscopy.Levels of IL-1β and IL-18 in cell supernatants were measured by enzyme-linked immunosorbent assay(ELISA).Protein expression of Nephrin,Podocin,PODXL,WT-1,Desmin,NLRP3,Cleaved Caspase-1,and GSDMD-N was detected via Western blot(WB).Immunofluorescence was used to assess expression levels of Nephrin,NLRP3,GSDMD-N,and Cleaved Caspase-1.Results Compared with the normal control group,the high glucose model group exhibited disrupted cell membrane integrity and reduced cell density.ELISA results showed upregulated expression of IL-1β and IL-18(P<0.05).Western blot analysis revealed decreased expression of Nephrin,Podocin,PODXL,and WT-1(P<0.05),and increased expression of Desmin,NLRP3,Cleaved Caspase-1,and GSDMD-N(P<0.05).Immu-nofluorescence indicated elevated relative fluorescence intensity of NLRP3,Cleaved Caspase-1,and GSDMD-N(P<0.05),alongside reduced Nephrin expression(P<0.05).Both the SQDH-containing serum group and MCC950 group showed significant amelioration of these abnormalities compared to the model group(P<0.05).Conclusion Serum containing SQDH modified prescription alleviates high glucose-induced podocyte injury,potentially by mitigating pyroptosis.
