基于氧化应激及PI3K/AKT通路的补气活血益精方改善肾虚血瘀型少弱精子症大鼠睾丸组织的研究OA
Investigating the mechanism of Buqi Huoxue Yijing Formula(补气活血益精方)on testicular tissue in oligoasthenospermia rats with kidney deficiency and blood stasis type based on oxida-tive stress and the PI3K/AKT pathway
目的 探讨补气活血益精方对肾虚血瘀型少弱精子症模型大鼠睾丸氧化应激损伤的改善作用及其对胞內磷脂酰肌醇激酶(PI3K)/蛋白激酶B(AKT)信号通路的调控机制.方法 选取SD雄性大鼠60只,随机分为空白组10只,造模组50只.采用雷公藤多苷40 mg·(kg·d)-1灌胃建立模型,造模成功后分为模型组、左卡尼汀组1.8 mL·(kg·d)-1及补气活血益精方低[7.87g·(kg·d)-1]、中[15.75g·(kg·d)-1]、高[31.5 g·(kg·d)-1]剂量组,持续治疗28d.实验8周后摘取双侧睾丸组织,观察指标包括:大鼠一般情况;全自动精子分析仪检测精子质量;切片苏木素-伊红(HE)染色法观察大鼠睾丸组织病理形态学改变;酶联免疫吸附测定法(ELISA)检测睾丸组织活性氧(ROS)、丙二醛(MDA)的含量;蛋白免疫印迹法(Western blot)检测PI3K、P-PI3K、AKT、P-AKT等蛋白的表达.结果 与空白组相比,模型组大鼠精子浓度及活力显著降低,睾丸组织出现曲细精管结构破坏,ROS和MDA水平显著升高(P<0.01),PI3K、p-PI3K、AKT、p-AKT蛋白表达显著降低(P<0.01).各治疗组(尤其高剂量组)能有效恢复生精小管结构,显著降低ROS和MDA水平(P<0.01,P<0.05),除低剂量组外,余治疗组均可显著上调PI3K/AKT通路相关蛋白表达(P<0.01).结论 补气活血益精方可能通过调控氧化应激及激活PI3K/AKT通路改善睾丸生精功能.
Objective To investigate the ameliorative effect of Buqi Huoxue Yijing Formula(补气活血益精方,BHYF)on oxidative stress damage in the testes of rats with kidney deficiency and blood stasis-type oligoasthenospermia(OAS)and its regulatory mechanism on the phosphoinositide 3-kinase(PI3K)/protein kinase B(AKT)signaling pathway.Methods Sixty male SD rats were randomly divided into a blank group(n=10)and a modeling group(n=50).The model was established by intragastric administration of Tripter-ygium glycosides(40 mg·kg-1·d-1).After successful modeling,the rats were further divided into the model group,L-carnitine group(1.8 mL·kg-1·d-1),and low/medium/high-dose BHYF groups(7.87,15.74,and 31.5 g·kg-1·d-1,respectively).Treatments contin-ued for 28 days.At the end of the 8-week experimental period,bilateral testicular tissues were harvested.The following indicators were assessed:the general condition of rats;sperm quality using an automated sperm analyzer;histopathological changes in testicular tissue by hematoxylin-eosin(HE)staining;levels of reactive oxygen species(ROS)and malondialdehyde(MDA)in testicular tissue by enzyme-linked immunosorbent assay(ELISA);and protein expression of PI3K,p-PI3K,AKT,and p-AKT by Western blot(WB).Results Compared with the blank group,the model group exhibited significantly reduced sperm concentration and motility,structural disruption of seminiferous tubules,markedly elevated levels of ROS and MDA(P<0.01),and significantly decreased expression of PI3K,p-PI3K,AKT,and p-AKT proteins(P<0.01).All treatment groups(especially the high-dose group)effectively restored semi-niferous tubule structure and significantly reduced ROS and MDA levels(P<0.01,P<0.05).Except for the low-dose group,all other treatment groups significantly upregulated the expression of PI3K/AKT pathway-related proteins(P<0.01).Conclusion BHYF may improve testicular spermatogenic function by regulating oxidative stress and activating the PI3K/AKT signaling pathway.
