目的 观察丹参红花配伍对脑缺血再灌注损伤(CIRI)的作用及与P-Smad3蛋白的相关性.方法 体外采用hCMEC/d3细胞建立OGD/R模型.分为Control、OGD/R、DH、SIS3 Hcl及SIS3 Hcl+DH组,检测细胞存活率、乳酸脱氢酶(LDH)释放、P-Smad3含量及细胞迁移率.体内以SD大鼠建立MCAO模型,分为Sham、MCAO、DH、SIS3 Hcl、SIS3 Hcl+DH及Edaravone组,行HE染色、免疫组化及Western blot检测脑组织形态与P-Smad3表达.结果 最佳建模条件为OGD 6h及85∼680 µmol/L药物浓度.与OGD/R组比较,DH组细胞存活率及迁移率显著升高,LDH释放下降,P-Smad3表达上升;SIS3 Hcl组则相反;SIS3 Hcl+DH组作用与DH组相似.动物实验结果一致:MCAO大鼠出现神经元减少、核固缩及P-Smad3上升,DH和Edara-vone可改善病理损伤并进一步促进P-Smad3表达,SIS3 Hcl加重损伤并抑制P-Smad3,联合DH后作用逆转.结论 丹参红花配伍可减轻CIRI细胞损伤、改善迁移率,机制可能与负调控P-Smad3通路相关.

Objective To investigate the protective effects of the Radix Salviae Miltiorrhizae and Flos Carthami medicinal couplet on cerebral ischemia-reperfusion injury(CIRI)and its association with P-Smad3.Methods An in vitro oxygen-glucose deprivation/reoxy-genation(OGD/R)model was established in hCMEC/d3 cells,which were divided into Control,OGD/R,DH,SIS3 HCl,and SIS3 HCl+DH groups.Cell viability,lactate dehydrogenase(LDH)release,P-Smad3 expression,and migration ability were assessed.In vivo,a middle cerebral artery occlusion(MCAO)model was induced in SD rats,which were randomized into Sham,MCAO,DH,SIS3 HCl,SIS3 HCl+DH,and Edaravone groups.Brain tissues were examined by hematoxylin-eosin staining(HE),immunohistochemistry(IHC),and Western blot(WB).Results Optimal modeling conditions were OGD for 6 h and drug concentrations of 85-680 µmol/L.Compared with the OGD/R group,the DH group showed significantly increased cell viability and migration,reduced LDH release,and elevated P-Smad3 expression,whereas SIS3 HCl exerted the opposite effects.The combination of SIS3 HCl with DH reversed the effects of SIS3 HCl alone.Consistent results were observed in vivo:MCAO rats exhibited neuronal loss,nuclear pyknosis,and elevated P-Smad3,which were alleviated by DH and Edaravone with further enhancement of P-Smad3.SIS3 HCl aggravated pathological damage and suppressed P-Smad3,while its combination with DH partially restored protective effects.Conclusion The Radix Salviae Miltiorrhi-zae and Flos Carthami medicinal couplet attenuates CIRI-induced cellular injury and enhances cell migration,potentially through nega-tive regulation of the P-Smad3 signaling pathway.
