分析感染传染性脾肾坏死病毒(ISKNV)的养殖翘嘴鳜(Siniperca chuatsi)池塘的水质及微生物群落结构特征,为解析ISKNV暴发后池塘水质及菌群结构的动态变化提供参考.测定ISKNV暴发前后的池塘水质指标,并采用16S rDNA测序技术分析池塘水体微生物群落结构.结果表明:ISKNV暴发后,池塘水体碱度、氨氮、硝酸盐氮、总氮和总磷积累率升高,池塘自净能力降低,水体菌群结构改变;在翘嘴鳜养殖池塘水体中,主要优势菌门为变形菌门(Proteobacteria)、拟杆菌门(Bacteroidota)和厚壁菌门(Firmicutes)等;ISKNV暴发后,池塘水体微生物丰富度和多样性下降,变形菌门的相对丰度降低,作为污染指示菌的拟杆菌门相对丰度增加,热单胞菌属(Thermomonas)、芽殖杆菌属(Gemmobacter)和军团菌属(Legionella)等相对丰度显著增加.

Microorganisms in the aquatic environment are crucial to aquatic ecosystems,playing impor-tant roles in pond water quality,disease prevention,nutrient dynamics and energy cycling.In this study,we analyzed physicochemical factors and microbial community structure of the water body in mandarin fish(Siniperca chuatsi)aquaculture ponds under healthy conditions and during outbreaks of infectious spleen and kidney necrosis virus(ISKNV).Our aim was to understand the dynamic changes in water qual-ity and bacterial community structure following an ISKNV outbreak From July to September of 2022,wa-ter samples were collected from two aquaculture ponds with healthy or infected fish.Water quality indica-tors for the two ponds before and after the ISKNV outbreak were determined and microbial community structure was determined using 16S rDNA sequencing technology.Results showed increased accumula-tion rates of alkalinity,ammonia nitrogen,nitrate nitrogen,total nitrogen,and total phosphorus in pond water after the ISKNV outbreak.The ISKNV outbreak decreased the self-purification capacity of the pond and changed the microbial community structure.The dominant bacterial phyla in the healthy S.ch-uatsi pond were Proteobacteria,Bacteroidetes,and Firmicutes.The ISKNV outbreak decreased microbial richness and diversity in the pond,decreased the relative abundance of Proteobacteria and increased the relative abundance of Bacteroidetes(an indicator of pollution)and significantly increased the relative abundances of the genera Thermomonas,Gemmobacter,and Legionella.Pearson correlation analysis indi-cated that genera such as Candidatus Aquiluna,the CL500-29 marine group,hgcl clade,Stenotrophomon-as,Cyanobium PCC-6307,and unclassified Illumatobacteraceae could potentially act as nitrogen-removal probiotics in the mandarin fish culture ponds and play a role in regulating water quality in aquaculture.
