目的:探讨加味桂枝茯苓汤(JGFD)通过PTEN诱导假定激酶1/帕金森病蛋白(PINK1/parkin)信号通路对高糖诱导的大鼠雪旺细胞(RSC96)损伤保护作用机制.方法:高糖诱导RSC96损伤,筛选确定50 mmol/L为高糖浓度;细胞计数套件(CCK-8)筛选出15%JGFD含药血清为最佳浓度.实验分为正常组、模型组(50 mmol/L葡萄糖+15%空白血清)、中药组(50 mmol/L葡萄糖+15%JGFD含药血清)、siRNA-PINK1组(加siRNA-PINK1转染).蛋白质印迹法验证PINK1转染效率,透射电镜观察线粒体及自噬结构,蛋白质印迹法和实时荧光定量PCR(RT-qPCR)检测PINK1、Parkin、P62、Beclin-1、微管相关蛋白1轻链3(LC3)的蛋白以及mRNA水平,流式细胞术检测活性氧(ROS).结果:高糖显著损伤RSC96,50 mmol/L时细胞存活较多;15%JGFD含药血清保护效果最佳.与正常组比较,模型组ROS升高(P<0.01),PINK1、Par-kin、Beclin-1、LC3表达降低,P62升高(P<0.01),细胞损伤严重且无自噬结构.与模型组比较,中药组ROS下降(P<0.01),上述分子表达趋势逆转(P<0.05或P<0.01),细胞损伤减轻且自噬结构恢复.siRNA-PINK1组则逆转中药组的保护作用(P<0.01),细胞损伤加重且自噬结构缺失.结论:JGFD能通过PINK1/Parkin信号通路介导线粒体自噬减轻RSC96细胞损伤,具有保护作用.

Objective:To investigate the protective mechanisms of Jiawei Guizhi Fuling Decoction(JGFD)against high glucose-in-duced injury in rat Schwann cells(RSC96)through the PTEN-induced putative kinase 1/Parkin(PINK1/Parkin)signaling path-way.Methods:RSC96 cell injury was induced by high glucose,and 50 mmol/L glucose was selected as the optimal concentration.Cell Counting Kit-8(CCK-8)assay identified 15%JGFD-containing serum as the optimal concentration.Cells were divided into the normal group,model group(50 mmol/L glucose+15%blank serum),JGFD group(50 mmol/L glucose+15%JGFD-containing serum),and siRNA-PINK1 group(siRNA-PINK1 transfection).PINK1 transfection efficiency was verified by Western blot.Mito-chondrial morphology and autophagic structures were observed by transmission electron microscopy.The protein and mRNA expres-sion levels of PINK1,Parkin,P62,Beclin-1,and microtubule-associated protein 1 light chain 3(LC3)were determined by Western blot and real-time quantitative PCR(RT-qPCR),respectively.Reactive oxygen species(ROS)levels were measured by flow cytom-etry.Results:High glucose significantly injured RSC96 cells,and 50 mmol/L glucose resulted in relatively higher cell viability.The protective effect of 15%JGFD-containing serum was optimal.Compared with the normal group,ROS levels were significantly in-creased in the model group(P<0.01),whereas the expression levels of PINK1,Parkin,Beclin-1,and LC3 were significantly de-creased and P62 expression was significantly increased(P<0.01).Severe cellular damage and the absence of autophagic structures were also observed.Compared with the model group,the JGFD group exhibited significantly reduced ROS levels(P<0.01),rever-sal of the above molecular expression changes(P<0.05 or P<0.01),alleviated cellular damage,and restoration of autophagic structures.In contrast,the siRNA-PINK1 group reversed the protective effects of JGFD(P<0.01),accompanied by aggravated cel-lular damage and loss of autophagic structures.Conclusion:JGFD exerts a protective effect against RSC96 cell injury by alleviating cellular damage through PINK1/Parkin signaling pathway-mediated mitophagy.
