骨乐冲剂抑制成骨细胞程序性坏死机制研究OACHSSCD
Mechanisms of Gule Infusion Granules in Suppression of Necroptosis of Osteoblasts
目的:探讨骨乐冲剂抑制成骨细胞程序性坏死的作用机制.方法:从乳鼠头盖骨中分离成骨细胞,并使用酯蟾毒配基(RBG)处理成骨细胞建立成骨细胞程序性坏死的细胞模型,将成骨细胞分成未经干预的对照组、模型组(20 μmol/L)以及在模型组基础上加入高剂量含药血清的骨乐冲剂组,对比分析各组细胞活性,碱性磷酸酶(ALP)活性,细胞内超微结,细胞线粒体膜通透性转换孔(MPTP)开放程度,细胞活性氧(ROS)水平,以及受体相互作用丝氨酸/苏氨酸蛋白激酶1(RIPK1)/受体相互作用丝氨酸/苏氨酸蛋白激酶3(RIPK3)/混合谱系激酶结构域样蛋白(MLKL)信号通路相关蛋白.结果:与对照组比较,RBG模型组的成骨细胞活力与ALP活性均显著降低;ROS水平、MPTP开放程度以及程序性坏死关键蛋白RIPK1、RIPK3和MLKL的表达均显著升高.骨乐冲剂组可显著逆转上述指标异常.透射电镜结果进一步显示,模型组细胞呈现典型的程序性坏死超微结构特征,而骨乐冲剂组能明显改善此类形态学改变.结论:骨乐冲剂可抑制成骨细胞程序性坏死,其机制可能与抑制RIPK1/RIPK3/MLKL信号通路有关,从而促进成骨细胞分化.
Objective:To investigate the mechanisms by which Gule Infusion Granules inhibit necroptosis in osteoblasts.Methods:Osteoblasts were isolated from the calvaria of neonatal rats,and a cellular model of osteoblast necroptosis was established by treat-ment with resibufogenin(RBG).The cells were divided into a control group without intervention,a model group treated with 20 μmol/L RBG,and a Gule Infusion Granules group treated with 20 μmol/L RBG in combination with high-dose Gule Infusion Granules-containing serum.Cell viability,alkaline phosphatase(ALP)activity,intracellular ultrastructural changes,mitochondrial permeability transition pore(MPTP)opening,reactive oxygen species(ROS)levels,and the expression of proteins associated with the RIPK1/RIPK3/MLKL signaling pathway were evaluated and compared among the groups.Results:Compared with the control group,osteoblasts in the RBG model group exhibited significantly reduced cell viability and ALP activity.In contrast,ROS levels,MPTP opening,and the expression levels of the key necroptosis-related proteins RIPK1,RIPK3,and MLKL were significantly in-creased.Treatment with Gule Infusion Granules significantly reversed these changes.Transmission electron microscopy further dem-onstrated that cells in the model group displayed typical ultrastructural features of necroptosis,whereas these morphological altera-tions were markedly improved in the Gule Infusion Granules group.Conclusion:Gule Infusion Granules inhibits osteoblast necrop-tosis and promotes osteoblast differentiation by activating the RIPK1/RIPK3/MLKL signaling pathway.
