基于指纹图谱与化学模式识别的多基原威灵仙质量评价研究OACHSSCD
Quality Evaluation of Clematidis Radix et Rhizoma from Multiple Sources Based on Fingerprint Profiles and Chemical Pattern Recognition
目的:建立不同基原威灵仙药材的指纹图谱,探讨威灵仙药材的质量差异及分类特征,为威灵仙药材的质量评价提供实验依据.方法:采用高效液相色谱(HPLC)法建立不同基源的威灵仙指纹图谱,通过相似度评价研究,结合聚类分析(CA)、主成分分析(PCA)和正交偏最小二乘法判别分析(OPLS-DA)统计方法,开展26批药材分类及主要影响成分研究.采用超高效液相色谱串联四极杆飞行时间质谱(UPLC-Q/TOF-MS)技术,对威灵仙药材的化学成分进行解析.结果:建立不同基原26个产地威灵仙药材指纹图谱,确定了 22个指纹峰,以8号峰常春藤皂苷元为基准物质,其相似度在0.219~1.000之间.CA、PCA、OPLS-DA将26批威灵仙药材均分为4类,并筛选出影响分类的特征成分7个,分别为峰14、峰19、峰11、峰22、峰12、峰8、峰13等7个色谱峰.成分解析出大豆黄酮、木兰花碱、橙皮素、川陈皮素等32个成分.结论:不同基原的威灵仙药材具有显著的成分差异特征,其特征与基原、产地均有较大关系,该研究为威灵仙质量评价提供科学依据.
Objective:To establish fingerprint profiles of Clematidis Radix et Rhizoma derived from different botanical origins,in-vestigate their quality differences and classification characteristics,and provide an experimental basis for the quality evaluation of Clematidis Radix et Rhizoma.Methods:High-performance liquid chromatography(HPLC)was employed to establish fingerprint profiles of Clematidis Radix et Rhizoma from different botanical origins.Similarity evaluation was performed,and cluster analysis(CA),principal component analysis(PCA),and orthogonal partial least squares discriminant analysis(OPLS-DA)were applied to classify 26 batches of medicinal materials and identify the major components contributing to their classification.Ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UPLC-Q/TOF-MS)was used to characterize the chemical constituents of Clematidis Radix et Rhizoma.Results:Fingerprint profiles were established for 26 batches of Clematidis Radix et Rhizoma collected from different production areas and representing different botanical origins.A total of 22 common finger-print peaks were identified.Using peak 8(hederagenin)as the reference peak,the similarity values ranged from 0.219 to 1.000.CA,PCA,and OPLS-DA consistently classified the 26 batches into four groups.Seven characteristic chromatographic peaks,namely peaks 14,19,11,22,12,8,and 13,were identified as the major contributors to the classification.UPLC-Q/TOF-MS analysis identi-fied 32 compounds,including daidzein,magnoflorine,hesperetin,and nobiletin.Conclusion:Clematidis Radix et Rhizoma from dif-ferent botanical origins exhibits significant differences in chemical composition.These characteristics are closely associated with both botanical origin and production area.This study provides a scientific basis for the quality evaluation of Clematidis Radix et Rhizoma.
