目的:基于网络药理学和动物实验的方法探讨黄芪赤风汤治疗脑梗死的分子机制.方法:采用网络药理学方法筛选出黄芪赤风汤有效成分及其与脑梗死的交集靶点等信息,应用分子对接技术验证主要有效成分与核心靶点对接情况,并用动物研究进行验证.结果:共获得56个活性成分,298个靶点.分子对接结果显示5个关键化合物和7个核心靶点能够直接相互作用,核心成分与核心蛋白具有较好的结合能.京都基因与基因组百科全书(KEGG)数据库的富集结果表明,黄芪赤风汤治疗脑梗死可能主要通过磷脂酰肌醇3激酶/蛋白激酶B(PI3K/AKT1)信号通路、肿瘤坏死因子(TNF)信号通路、白细胞介素17(IL-17)信号通路、血脂与动脉粥样硬化、晚期糖基化终末产物-晚期糖基化终末产物受体(AGE-RAGE)信号通路、流体剪切应力与动脉粥样硬化等信号通路等发挥作用.实验验证发现黄芪赤风汤能改善大鼠神经功能缺损,降低大鼠外周血清中IL-6、TNF、IL-1β水平,改善大鼠脑组织AKT1、TNF的表达.结论:黄芪赤风汤治疗脑梗死与调控AKT1、TNF、IL-6、IL-1β等核心靶点有关.

Objective:To investigate the molecular mechanisms underlying the therapeutic effects of Huangqi Chifeng Decoction on cerebral infarction using network pharmacology and animal experiments.Methods:Network pharmacology was employed to identify the active components of Huangqi Chifeng Decoction and their potential targets associated with cerebral infarction.Molecular doc-king was performed to verify the interactions between the major active compounds and core targets,and the findings were further validated through animal experiments.Results:A total of 56 active compounds and 298 targets were identified.Molecular docking a-nalysis demonstrated that five key compounds could directly interact with seven core targets,and the core compounds exhibited fa-vorable binding affinities with the core proteins.Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis indicated that Huangqi Chifeng Decoction may exert therapeutic effects on cerebral infarction primarily through the phosphoinositide 3-kinase/protein kinase B(PI3K/AKT1)signaling pathway,tumor necrosis factor(TNF)signaling pathway,interleukin-17(IL-17)signaling pathway,lipid and atherosclerosis pathway,advanced glycation end product-receptor for advanced glycation end product(AGE-RAGE)signaling pathway,and fluid shear stress and atherosclerosis pathway.Experimental validation demonstrated that Huangqi Chifeng Decoction improved neurological deficits in rats,reduced the levels of IL-6,TNF,and IL-1β in peripheral serum,and improved the expression of AKT1 and TNF in brain tissue.Conclusion:The therapeutic effects of Huangqi Chifeng Decoction on cerebral infarction may be associated with the regulation of core targets,including AKT1,TNF,IL-6,and IL-1β.
