凤香洗液对子宫内膜癌细胞的影响及机制研究OACHSSCD

Effects and Mechanisms of Fengxiang Lotion on Endometrial Cancer Cells

目的:探讨凤香洗液通过丝裂原活化蛋白激酶(MAPK)信号通路对子宫内膜癌HEC-1B细胞增殖、凋亡和自噬的影响及其作用机制.方法:制备凤香提取液,不同浓度凤香提取液作用HEC-1B细胞,采用噻唑蓝(MTT)法检测凤香提取液对HEC-1B细胞增殖的影响,4',6-二脒基-2-苯基吲哚染色法(DAPI)观察细胞形态,单丹磺酰戊二胺染色法(MDC)检测细胞自噬,流式细胞术检测细胞周期和凋亡,蛋白质印迹法(WB)检测细胞周期、自噬、凋亡和MAPK通路相关蛋白的表达.结果:凤香提取液对HEC-1B细胞增殖的抑制作用呈现明显的浓度和时间依赖性,HEC-1B细胞经2 mg/mL凤香提取液处理48 h后,与0 mg/mL组比较,G0/G1期细胞比例降低(P<0.01),S期和G2/M期细胞比例升高(P<0.01);随着药物浓度增加,细胞周期相关蛋白周期蛋白A(Cyclin A)、Cyclin依赖性激酶2(CDK2)表达逐渐下降,凤香提取液显著抑制HEC-1B细胞增殖,使细胞体积缩小、细胞核皱缩;细胞凋亡率逐渐上升,抗凋亡蛋白B淋巴细胞瘤2(Bcl-2)表达逐渐下降,凋亡蛋白Cleaved胱天蛋白酶-9(Cleaved Caspase-9)和促凋亡蛋白Bcl-2相关X蛋白(Bax)表达逐渐上升,凤香提取液显著促进细胞自噬的发生,且p62蛋白表达呈浓度依赖性降低,凤香提取液显著增加JNK和p38磷酸化水平.结论:凤香提取液通过抑制CDK2和Cyclin A蛋白表达,阻滞细胞周期,抑制细胞增殖,同时通过升高Jun激酶(JNK)和p38蛋白磷酸化水平,活化MAPK信号通路,诱导细胞凋亡和自噬.

Objective:To investigate the effects of Fengxiang Lotion on the proliferation,apoptosis,and autophagy of human endom-etrial cancer HEC-1B cells through the mitogen-activated protein kinase(MAPK)signaling pathway and to explore the underlying mechanisms.Methods:Fengxiang Lotion was prepared and applied to HEC-1B cells at different concentrations.Cell proliferation of HEC-1B cells was assessed using the methyl thiazolyl tetrazolium(MTT)assay.Cellular morphology was observed by 4',6-diamidi-no-2-phenylindole(DAPI)staining.Autophagy was evaluated by monodansylcadaverine(MDC)staining.Cell cycle distribution and apoptosis were analyzed by flow cytometry.Western blot analysis was performed to determine the expression levels of proteins asso-ciated with the cell cycle,autophagy,apoptosis,and the MAPK signaling pathway.Results:Fengxiang Lotion inhibited the prolifera-tion of HEC-1B cells in a concentration-and time-dependent manner.After treatment with 2 mg/mL Fengxiang Lotion for 48 h,the proportion of cells in the G0/G1 phase was decreased,whereas the proportions of cells in the S and G2/M phases were increased compared with those in the 0 mg/mL group(P<0.01).With increasing concentrations of Fengxiang Lotion,the expression levels of the cell cycle-related proteins Cyclin A and cyclin-dependent kinase 2(CDK2)gradually decreased.Fengxiang Lotion significantly inhibited HEC-1B cell proliferation,accompanied by reduced cell volume and nuclear shrinkage.The apoptotic rate increased pro-gressively,while the expression of the anti-apoptotic protein B-cell lymphoma 2(Bcl-2)gradually decreased.In contrast,the expres-sion levels of cleaved caspase-9 and the pro-apoptotic protein Bcl-2-associated X protein(Bax)gradually increased.Fengxiang Lo-tion markedly promoted autophagy,and p62 protein expression decreased in a concentration-dependent manner.In addition,Fengx-iang Lotion significantly increased the phosphorylation levels of c-Jun N-terminal kinase(JNK)and p38.Conclusion:Fengxiang Lotion inhibits the proliferation of HEC-1B cells by suppressing the expression of CDK2 and Cyclin A,thereby inducing cell-cycle arrest.Meanwhile,it increases the phosphorylation levels of JNK and p38,activates the MAPK signaling pathway,and induces apop-tosis and autophagy.
