四君子汤对高压电烧伤大鼠免疫功能及血管内皮生长因子表达的影响OACHSSCD
Effects of Sijunzi Decoction on Immune Function and Expression of Vascular Endothelial Growth Factor in High-voltage Electrical Burn Rats
目的:探讨四君子汤对高压电烧伤大鼠免疫功能及血管生长因子的影响.方法:将120只健康雄性SD大鼠按照简单随机方法分为假伤组、模型组、四君子汤低剂量组、四君子汤高剂量组,每组30只.假伤组大鼠不进行电击,其余组大鼠均构建高压电烧伤模型.建模成功后,四君子汤低、高剂量组大鼠分别灌胃2.86 g/kg、5.72 g/kg四君子汤提取液,假伤组、模型组灌胃等体积生理盐水.于建模后48 h、72 h、7 d,采集血样及免疫器官备用,用试剂盒法检测血清中免疫球蛋白(Ig)G、IgA、IgM、炎症介质、血管内皮生长因子(VEGF)及血小板活化因子(PAF)、血小板生成素(TPO)水平;逆转录定量聚合酶链式反应法检测各组大鼠血清STAT3 mRNA、VEGF mRNA表达水平.结果:与假伤组比较,模型组胸腺、脾脏指数及血清IgG、IgA、IgM水平均显著降低,血清促炎症介质、VEGF、PAF、TPO水平均显著升高(P<0.05);与模型组比较,四君子汤低、高剂量组大鼠胸腺、脾脏指数及血清IgG、IgA、IgM、VEGF水平均显著升高,血清促炎症介质、PAF、TPO水平均显著下降(P<0.05).结论:四君子汤可通过介导STAT3上调VEGF表达以加速愈合,抑制PAF、TPO水平以改善血管损伤,同时改善免疫功能障碍,进而减轻高压电烧伤.
Objective:To investigate the effects of Sijunzi Decoction on immune function and vascular growth factors in rats with high-voltage electrical burns.Methods:A total of 120 healthy male SD rats were randomly assigned into four groups(n=30 each):sham injury group,model group,low-dose Sijunzi Decoction group,and high-dose Sijunzi Decoction group.Rats in the sham injury group did not receive electric shock,while a high-voltage electrical burn model was established in the other groups.After modeling,rats in the low-and high-dose groups received Sijunzi Decoction extract via gavage at 2.86 g/kg and 5.72 g/kg,respectively,while the sham injury and model groups received an equal volume of saline.Blood samples and immune organs were collected at 48 h,72 h,and 7 d after modeling.Serum immunoglobulins(IgG,IgA,IgM),inflammatory cytokines,vascular endothelial growth factor(VEGF),platelet-activating factor(PAF),and thrombopoietin(TPO)were measured using commercial kits.STAT3 mRNA and VEGF mRNA expression levels in serum were determined by reverse transcription quantitative PCR.Results:Compared with the sham injury group,the model group exhibited significantly decreased thymus and spleen indices and lower serum IgG,IgA,and IgM levels,along with significantly elevated serum pro-inflammatory cytokines,VEGF,PAF,and TPO levels(P<0.05).Compared with the model group,rats in the low-and high-dose Sijunzi Decoction groups showed significant increases in thymus and spleen indices and serum IgG,IgA,IgM,and VEGF levels,and significant decreases in pro-inflammatory cytokines,PAF,and TPO levels(P<0.05).Conclusion:Sijunzi Decoction may alleviate high-voltage electrical burn injury by upregulating STAT3 to enhance VEGF expression and promote healing,inhibiting PAF and TPO to reduce vascular injury,and improving immune function.
