参萸养心胶囊对心肌纤维化小鼠的影响及作用机制OACHSSCD
Effects and Mechanisms of Shenyu Yangxin Capsules on Myocardial Fibrosis in Mice
目的:评估参萸养心胶囊对心肌纤维化(MF)的疗效、作用靶点及作用机制.方法:50只雄性C57BL/6J小鼠按随机数字表分为对照组、模型组、参萸养心胶囊低、高(胶囊内容物冻干粉1 mg/d、2 mg/d)剂量组和卡托普利组(0.2 mg/d),每组10只.除对照组外,其余各组小鼠予以异丙肾上腺素(5 mg/kg)皮下多点注射,1次/d,连续14 d以建立MF模型.造模结束当日,参萸养心胶囊低、高剂量组和卡托普利组进行灌胃给药,对照组和模型组给予等体积蒸馏水.给药14 d后,通过超声检测小鼠左室射血分数(LVEF)、左室短轴缩短率(LVFS);马松(Masson)染色评估小鼠MF程度;免疫组化法检测小鼠心肌组织中转化生长因子-β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)、Ⅲ型胶原蛋白(COLⅢ)蛋白表达;Re-al-time PCR检测小鼠心肌组织中白细胞介素-6(IL-6)、转化生长因子-β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)、ColⅢmRNA表达;蛋白质印迹法检测小鼠心肌组织TGF-β1、α-SMA、Smad3、p-Smad3蛋白表达.结果:与对照组比较,模型组小鼠LVEF、LVFS明显降低,心肌胶原面积分数、胶原阳性面积明显升高(P<0.01),IL-6、TGF-β1、α-SMA、Col Ⅲ mRNA表达显著升高(P<0.01),TGF-β1、α-SMA、Smad3、p-Smad3蛋白表达显著升高(P<0.01).与模型组比较,卡托普利组及参萸养心胶囊低、高剂量组上述指标均得到不同程度的改善,差异均有统计学意义(均P<0.01).结论:参萸养心胶囊可改善异丙肾上腺素诱导的小鼠心功能障碍,减少胶原蛋白沉积,延缓MF病理进程,其机制可能与抑制TGF-β1/Smad3信号通路来改善异丙肾上腺素诱导的小鼠MF有关.
Objective:To evaluate the efficacy,targets,and mechanisms of Shenyu Yangxin Capsules on myocardial fibrosis(MF)in mice.Methods:Fifty male C57BL/6J mice were randomly assigned to five groups(n=10 each)according to a random number table:control group,model group,low-and high-dose Shenyu Yangxin Capsules group(1 and 2 mg/d for capsule contents in freeze-dried powder),and captopril group(0.2 mg/d).Except for the control group,mice in all other groups were injected subcutaneously with isoproterenol(5 mg/kg)once daily for 14 days to induce MF.On the day of model establishment,the low-and high-dose She-nyu Yangxin Capsules groups and the captopril group received oral administration,while the control and model groups received an equal volume of distilled water.After 14 days of treatment,cardiac function was assessed by ultrasound,measuring left ventricular e-jection fraction(LVEF)and left ventricular fractional shortening(LVFS).MF was evaluated by Masson's trichrome staining.Immu-nohistochemistry was performed to detect protein expression of transforming growth factor-β1(TGF-β1),alpha-smooth muscle actin(α-SMA),and collagen Ⅲ(Col Ⅲ)in myocardial tissue.Real-time PCR was used to measure mRNA expression of interleukin-6(IL-6),TGF-β1,α-SMA,and Col Ⅲ.Western blot analysis was performed to detect protein levels of TGF-β1,α-SMA,Smad3,and phospho-Smad3(p-Smad3).Results:Compared with the control group,mice in the model group exhibited significantly reduced LVEF and LVFS,increased myocardial collagen volume fraction and collagen-positive area(P<0.01),and significantly elevated mRNA expression of IL-6,TGF-β1,α-SMA,and Col Ⅲ(P<0.01),as well as protein levels of TGF-β1,α-SMA,Smad3,and p-Smad3(P<0.01).Compared with the model group,the captopril group and both Shenyu Yangxin Capsules groups showed varying degrees of improvement in all the above indicators,with all differences statistically significant(P<0.01).Conclusion:Shenyu Yangxin Capsules ameliorates isoproterenol-induced cardiac dysfunction,reduces collagen deposition,and delays the progression of MF in mice,potentially by inhibiting the TGF-β1/Smad3 signaling pathway.
