Effects of Herbal-Cake Partition Moxibustion on Fecal Extracellular Vesicles miRNAs in Atherosclerosis

目的 从动脉粥样硬化(Atherosclerosis,AS)兔粪便来源外囊泡(Extracellular vesicles,EVs)miRNA的角度揭示隔药饼灸防治AS的作用机制.方法 选取新西兰兔随机分为正常组、模型组和隔药饼灸组,后两组采用高脂饲料110-120 g·d-1,连续12周构建AS模型.隔药饼灸组选取A组:巨阙、天枢、丰隆;B组:心俞、肝俞、脾俞等穴位,每日1次,每穴灸4壮,两组穴位交替施灸,持续12周.干预结束后,检测血清血脂指标,观察主动脉组织病理变化(苏木精-伊红染色、油红O染色),并通过透射电镜、纳米粒径跟踪分析、Western blotting鉴定粪便来源EVs.同时对EVs进行miRNA测序,筛选差异表达miRNA,预测靶基因并进行基因本体(Gene ontology,GO)、京都基因和基因组数据库(Kyoto Encyclopedia of Genes and Genomes,KEGG)富集分析.结果 与模型组相比,隔药饼灸组总胆固醇(Total cholesterol,TC)、甘油三酯(Triglycerides,TG)、低密度脂蛋白胆固醇(Low-density lipoprotein cholesterol,LDL-C)显著降低,高密度脂蛋白胆固醇(High density lipoprotein cholesterol,HDL-C)升高(P<0.05),主动脉内皮增厚、泡沫细胞堆积明显改善,斑块面积减少.模型组与正常组比较有27个差异miRNA,隔药饼灸组与模型组比较有18个差异miRNA,取后者比较中表达显著且丰度较高的miRNA(miR-22-3p、miR-26b-5p、let-7a-5p)进行分析.GO分析中靶基因调控基因表达与蛋白质功能,涉及细胞凋亡或损伤修复、细胞黏附和细胞外基质重塑等.KEGG分析中这些通路显示靶基因覆盖细胞黏附、信号传导、基因表达调控、炎症反应、代谢调控等多个方面.结论 隔药饼灸能显著改善AS血脂情况及血管内皮结构,减少斑块形成,并引起粪便来源EVs miRNAs表达差异,为未来从EVs miRNAs以及肠-血管轴的角度研究AS的治疗机制奠定了基础.

Objective To investigate the mechanism by which herbal-cake partition moxibustion prevents and treats atherosclerosis(AS)by examining its regulatory effects on extracellular vesicles(EVs)miRNAs derived from colonic feces in AS rabbits.Methods New Zealand rabbits were randomly divided into a normal group,a model group,and a moxibustion group.The model and moxibustion groups were fed a high-fat diet(110-120 g·d-1)for 12 consecutive weeks to induce AS.The moxibustion group received treatment at two alternating sets of acupoints for 12 weeks:Set A:Juque(CV14),Tianshu(ST25),Fenglong(ST40);Set B:Xinshu(BL15),Ganshu(BL18),Pishu(BL20).Moxibustion was applied once daily,four moxa cones per acupoint.After the intervention,serum lipid profiles were assessed,and pathological changes in the aorta were examined using HE and Oil Red O staining.Fecal EVs were identified using transmission electron microscopy,nanoparticle tracking analysis,and Western blotting.EVs miRNAs were then sequenced,differentially expressed miRNAs were screened,and their target genes were predicted for GO and KEGG enrichment analysis.Results Therapeutic effects:Compared with the model group,the moxibustion group showed significantly lower TC,TG,and LDL-C levels and higher HDL-C levels(P<0.05).Endothelial thickening and foam cell accumulation in the aorta were markedly improved,and plaque area was reduced.miRNA expression:twenty-seven miRNAs were differentially expressed between the model and normal groups,and 18 miRNAs differed between the moxibustion and model groups.Among the latter,miRNAs with both significant expression changes and high abundance(miR-22-3p,miR-26b-5p,and let-7a-5p)were selected for further analysis.Target gene enrichment:GO analysis indicated that target genes were involved in the regulation of gene expression and protein function,including pathways related to apoptosis or tissue repair,cell adhesion,and extracellular matrix remodeling.KEGG analysis showed that enriched pathways were associated with cell adhesion,signal transduction,gene expression regulation,inflammatory responses,and metabolic regulation.Conclusion Herbal-cake partition moxibustion significantly improves lipid profiles and vascular endothelial structure in AS rabbits and reduces plaque formation.It also alters fecal EVs miRNA expression,providing a foundation for future studies on the treatment of AS from the perspective of EVs miRNAs and the gut-vascular axis.
