救必应抗炎活性评价及抗炎机制初探OA
Evaluation of anti-inflammatory activity and preliminary investigation of anti-inflammatory mechanism of Jiubiying
目的:基于斑马鱼炎症模型和网络药理学技术探讨救必应抗炎活性及其机制.方法:建立硫酸铜浸泡、断尾和脂多糖显微注射致斑马鱼炎症模型,观察中性粒细胞迁移聚集情况,进一步采用生存分析、病理形态观察和荧光定量聚合酶链式反应实验评价救必应的抗炎活性,最后结合网络药理学预测救必应抗炎作用的靶点和信号通路.结果:救必应可显著抑制硫酸铜浸泡、断尾和脂多糖显微注射致炎斑马鱼幼鱼中性粒细胞向炎症部位迁移聚集,提高脂多糖致炎斑马鱼的存活率,并可降低炎性因子白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的表达.网络药理学分析显示救必应主要通过救必应酸、冬青苷A、藏红花酸二甲酯和圆叶帽柱木碱作用于肿瘤坏死因子(TNF)、信号转导和转录激活因子3(STAT3)、基质金属蛋白酶2(MMP2)、表皮生长因子受体(EGFR)、丝氨酸/苏氨酸激酶1(NR3C1)等多个核心靶点发挥抗炎作用,主要涉及松弛素信号通路、癌症蛋白多糖通路及黏着斑通路等.结论:救必应通过多成分、多靶点、多通路机制发挥抗炎作用,为后续研究开发提供理论支撑.
Objective:To investigate the anti-inflammatory activity and mechanism of Jiubiying using zebrafish inflam-mation models and network pharmacology technology.Methods:Zebrafish inflammation models were established by copper sulfate immersion,tail amputation,and lipopolysaccharide microinjection.Neutrophil migration and aggregation were ob-served.Further evaluations of anti-inflammatory activity were performed using survival analysis,pathological morphology ob-servation,and quantitative real-time PCR.Finally,network pharmacology was used to predict the anti-inflammatory targets and signaling pathways of Jiubiying.Results:Jiubiying significantly inhibited neutrophil migration and aggregation toward inflammatory sites in zebrafish larvae induced by copper sulfate immersion,tail amputation,and lipopolysaccharide microin-jection.It improved the survival rate of lipopolysaccharide-inflamed zebrafish and reduced the expression of inflammatory cytokines interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α).Network pharmacology analysis indicated that Jiubiy-ing exerted its anti-inflammatory effects mainly through components such as Jiubiying acid,ilexin A,crocetin dimethyl es-ter,and rotundifoline,acting on multiple core targets including tumor necrosis factor(TNF),signal transducer and activator of transcription 3(STAT3),matrix metalloproteinase 2(MMP2),epidermal growth factor receptor(EGFR),and serine/threonine kinase 1(NR3C1),primarily involving the relaxin signaling pathway,proteoglycans in cancer pathway,and focal adhesion pathway.Conclusion:Jiubiying exerts anti-inflammatory effects through a multi-component,multi-target,and multi-pathway mechanism,providing theoretical support for further research and development.
