菌酶协同提升杏鲍菇多糖溶出量及抗氧化活性研究OA
Enhancement of Pleurotus eryngii Polysaccharide Extraction Amount and Antioxidant Activity by Microbial-Enzyme Synergy
本研究以杏鲍菇为原料,基于复合酶解与发酵协同处理,提升杏鲍菇多糖溶出量及抗氧化活性,为杏鲍菇的精深加工提供理论依据.在筛选酶的种类及复合酶最佳配比(纤维素酶、木瓜蛋白酶、木聚糖酶质量比为3∶3∶2)的基础上,分别以杏鲍菇多糖溶出量、DPPH自由基清除率为指标,采用单因素试验及响应面法优化复合酶解及发酵工艺,并对二者的协同作用方式进行筛选.结果表明,在复合酶添加量为4.0%(按杏鲍菇粉质量计)、50 ℃、pH 5.0条件下酶解2 h,多糖溶出量最高,达15.07 mg/mL;在发酵菌粉添加量为1.0%、料液比1∶20条件下发酵12 h,杏鲍菇发酵液的DPPH自由基清除率为94.10%.在先酶解后发酵(加pH 5.0缓冲液)的协同方式下,杏鲍菇多糖溶出量达到17.42 mg/mL,DPPH自由基清除能力达到96.14%,与单独酶解最优条件下相比多糖溶出量提高15.59%,与单独发酵最优条件相比DPPH自由基清除率提高2.17%.由此可见,酶解和发酵协同作用可以提高杏鲍菇多糖溶出量和抗氧化能力.
This study aimed to enhance the extraction amount and antioxidant activity of polysaccharides from Pleurotus eryngii through a synergistic process combining complex enzymatic hydrolysis and fermentation,thereby providing theoretical foundations for the deep processing of P.eryngii.On the basis of screening en-zyme types and optimizing the optimal mass ratio of enzyme complex(cellulosease,papain and xylanase at the ratio of 3∶3∶2),single factor tests and response surface methodology were employed to optimize the processes and screen the synergistic interaction modes of enzymatic hydrolysis and fermentation using polysaccharide ex-traction amount and DPPH radical scavenging rate as evaluation indices.The results demonstrated that when enzymatic hydrolysis for 2 h at 50 ℃ and pH 5.0 with 4.0%of enzyme complex dosage(by the mass of mush-room powder),the polysaccharide extraction amount reached 15.07 mg/mL.When fermentation for 12 h under the conditions of adding 1.0%lactic acid bacteria inoculum and the solid-to-liquid ratio of 1∶20,the DPPH radical scavenging rate of the fermented broth of P.eryngii was 94.10%.Under the synergistic mode of enzy-matic hydrolysis followed by fermentation(adding pH 5.0 buffer solution),the polysaccharide extraction a-mount of P.eryngii reached 17.42 mg/mL,and the DPPH radical scavenging capacity reached 96.14%,which increased by 15.59%and 2.17%compared with the optimal enzymatic hydrolysis process alone and the optimal fermentation process alone,respectively.These results demonstrated that the synergistic interaction of enzymatic hydrolysis and fermentation could significantly enhanced both polysaccharide extraction amount and antioxidant capacity of P.eryngii.
