桃PP2C基因家族全基因组鉴定及表达分析OA
Genome-Wide Identification and Expression Analysis of PP2C Gene Family in Peach(Prunus persica)
PP2C(2C型蛋白磷酸酶)是一种在生物体内普遍存在的蛋白磷酸酶,在植物应对逆境胁迫以及激素响应等生理过程中具有至关重要的作用.然而,目前关于PP2C在桃树(Prunus persica)中的研究尚显不足,有待进一步探索.本研究以'21世纪'桃树为试验材料,利用生物信息学方法从其全基因组中筛选出具有完整PP2C保守结构域的基因共56个,其蛋白氨基酸序列长度介于169~1 080 aa之间,分子量介于19.20~119.48 kDa之间.系统发育分析发现这56个PpPP2C蛋白可归为7个亚族.组织表达特异性分析结果揭示,PpPP2C家族基因在桃树的不同组织部位中呈现出不同的表达模式,PpPP2C40在茎中的表达量最高,Pp-PP2C2、PpPP2C9、PpPP2C14、PpPP2C26、PpPP2C30、PpPP2C48 和 PpPP2C51 在芽中的表达量最高,PpPP2C12和PpPP2C43在叶中的表达量最高.此外,对桃树一年生盆栽苗进行0 ℃低温处理,RT-qPCR分析发现2个PpPP2C基因上调、8个下调,表明该家族成员在冷响应中发生了功能分化.本研究首次系统鉴定了桃Pp-PP2C基因家族,揭示了芽和叶中特异表达的PpPP2C基因可能参与器官发育调控,并发现多数成员对低温胁迫敏感且表达受抑制,这为桃PP2C基因功能的深入研究提供了参考.
PP2C(Type 2C protein phosphatase)is a class of ubiquitously present protein phosphatases that play critical roles in various physiological processes in the plants,including stress response and hormone signaling.However,the research on PP2C in peach(Prunus persica)remains limited and requires further ex-ploration.In this study,using'21 Shiji'peach as experimental material,a total of 56 genes harboring an in-tact PP2C conserved domain were identified from the peach genome through bioinformatics methods.The length of their encoded proteins ranged from 169 to 1 080 amino acids,and the molecular weight of these pro-teins ranged from 19.20 kDa to 119.48 kDa.Phylogenetic analysis revealed that the 56 PpPP2C proteins could be classified into seven subfamilies.Tissue-specific expression analysis showed that PpPP2C genes exhibited distinct expression patterns across different peach tissues.PpPP2C40 was predominantly expressed in stems,PpPP2C2,PpPP2C9,PpPP2C14,PpPP2C26,PpPP2C30,PpPP2C48 and PpPP2C51 showed the highest expression levels in buds,and PpPP2C12 and PpPP2C43 were the most highly expressed in leaves.Further-more,one-year-old potted peach seedlings were subjected to 0 ℃ low-temperature treatment,and RT-qPCR a-nalysis revealed that the expression of two PpPP2C genes were up-regulated while that of eight genes were down-regulated,suggesting functional divergence among family members in response to cold.This study pro-vided the first systematic identification of peach PpPP2C gene family,revealed that certain genes specifically expressed in buds and leaves might be involved in organ development,and demonstrated that most members were cold-responsive with suppressed expression under low-temperature stress.These findings could provide a reference for future in-depth functional research of PP2C genes in peach.
