花生DOG1基因家族的鉴定与表达分析OA
Identification and Expression Analysis of DOG1 Gene Family in Peanut
DOG1(Delay of germination 1)基因是种子休眠的一个重要调控基因.为探索花生种子休眠机制,本研究从栽培种花生(Arachis hypogaea L.)全基因组中鉴定出35个DOG1基因家族成员,并对其编码蛋白理化性质、系统进化及基因表达模式进行分析.结果表明,DOG1基因家族成员编码的氨基酸数量在126~662 aa之间,分子量在15.122~75.166 kDa之间,理论等电点在5.16~8.79之间,约68.6%的蛋白为弱酸性蛋白,全部为亲水性蛋白;系统进化分析显示这35个基因分布于18条染色体上,可分为7个亚家族.DOG1基因具有组织表达特异性,选择在花生籽仁中表达量较高的4个基因(Ah0HHX3D、Ah5PG8C6、AhE6CWZU和AhDXWW21),利用荧光定量PCR(RT-qPCR)检测其在花生种质H7500和ZP06种子萌发1、3、5、7 d的表达量,结果显示,Ah0HHX3D和AhE6CWZU的表达量在萌发种子胚中呈现逐步降低的趋势,而在不发芽种子中则一直处于较高水平,证明其可能参与到种子萌发过程中.本研究结果可为进一步研究DOC1基因家族在花生种子萌发过程中的功能奠定基础.
The DOG1(Delay of germination 1)gene is an important regulatory gene controlling seed dormancy.In order to explore the mechanism of peanut seed dormancy,a total of 35 DOG1 gene family mem-bers were identified from the whole genome of cultivated peanut(Arachis hypogaea L.)in this study,and the physicochemical properties,phylogenetic evolution of their coding proteins and the gene expression patterns were analyzed.The results showed that the number of amino acids encoded by DOG1 gene family members ranged from 126 to 662;the DOG1 proteins had the molecular weight ranging from 15.122 to 75.166 kDa,the theoretical isoelectric point between 5.16 and 8.79,and about 68.6%of them were weakly acidic proteins,while all were hydrophilic proteins.Phylogenetic evolution analysis showed that the 35 DOG1 gene family members were distributed on 18 chromosomes and could be divided into 7 subfamilies.The DOG1 genes showed tissue expression specificity.Four genes(Ah0HHX3D,Ah5PG8C6,AhE6CWZU and AhDXWW21)with kernel-specific expression were selected for fluorescence quantitative PCR(RT-qPCR)analysis and their expression levels at 1,3,5 and 7 d of germination in H7500 and ZP06 peanut seeds were analyzed.The re-sults showed that the expression level of Ah0HHX3D and AhE6CWZU showed the gradually decreasing trend in the embryos of germinated seeds,but were always at a higher level in non-germinated seeds,proving that they might be involved in the process of seed germination.These results could lay a foundation for further research on the function of DOG1 gene family in seed germination process of peanut.
