甘蓝型油菜ABI3基因的克隆、表达分析和功能的初步研究OA
Cloning,Expression Analysis,and Preliminary Functional Research of the ABI3 Gene in Brassica napus
[目的]克隆甘蓝型油菜ABI3基因的CDS序列,对BnABI3基因结构及其编码的蛋白质性质和在逆境胁迫下的表达模式进行分析,为探索ABI3基因在响应非生物胁迫过程中的调控作用提供参考.[方法]以甘蓝型油菜"中油821"的叶片cDNA为模板,克隆获得BnABI3基因的CDS序列.对其进行生物信息学分析,利用qRT-PCR技术分析其在逆境胁迫下的表达,利用Y2H和BiFC实验验证蛋白互作情况.[结果]甘蓝型油菜BnABI3基因CDS区全长2 133 bp,共编码710个氨基酸,与拟南芥中AtABI3蛋白序列的同源一致性为84.33%;BnABI3基因能够响应干旱、低温和盐胁迫处理,且在盐胁迫下呈急剧诱导表达,响应强度高于其他胁迫处理;BnABI3蛋白定位于细胞核,酵母双杂交和双分子荧光互补分析表明其能够与转录因子BnABI5相互作用.[结论]BnABI3是核定位蛋白,可能通过与BnABI5互作介导BnABI5调控的生物学活动,参与甘蓝型油菜响应干旱、低温和盐胁迫过程.研究结果为揭示BnABI3抗逆性的调控机制提供参考,为后续深入研究BnABI3基因的功能奠定基础.
[Objective]To clone the coding sequence(CDS)of the Brassica napus ABI3 gene,analyze the structural features of the BnABI3 gene,the properties of its encoded protein,and its expression pat-tern under abiotic stress,thereby providing insights into the regulatory role of the ABI3 gene in respond-ing to abiotic stress.[Method]Using leaf cDNA from Brassica napus"Zhongyou 821"as a template,the BnABI3 gene CDS sequence was cloned.Bioinformatics analysis was performed,and its expression under abiotic stress was analyzed using qRT-PCR.Protein interactions were verified through Y2H and BiFC ex-periments.[Result]The CDS region of BnABI3 gene in B.napus was 2 133 bp in length,encoding 710 amino acids.The homology with AtABI3 protein sequence in A.thaliana was 84.33%.The BnABI3 gene responds to drought,low temperature,and salt stress treatments,exhibiting sharply induced expression under salt stress with a response intensity higher than other stress treatments.The BnABI3 protein local-ized to the nucleus,and yeast two-hybrid and bimolecular fluorescence complementation analyses indi-cated its interaction with the transcription factor BnABI5.[Conclusion]BnABI3 is a nuclear-localized protein that likely mediates BnABI5-regulated biological activities through interaction with BnABI5,par-ticipating in Brassica napus responses to drought,low temperature,and salt stress.These findings pro-vide insights into the regulatory mechanisms of BnABI3 stress resistance and lay the foundation for further functional studies of the BnABI3 gene.
