榴莲花粉活力测定方法的比较研究OA
Comparative Study on the Determination Methods of Pollen Viability of Durio zibethinus
以金枕和猫山王榴莲品种为试验材料,采集盛花期花粉,通过亚历山大染色法、MTT 染色法对保存 2、4、8、12、18、24 h 时的榴莲花粉活力进行测定,并采用离体培养萌发法(固体和液体离体培养萌发法)观察不同培养时间的花粉萌发率.结果表明:两品种的花粉活力均随保存时间延长呈下降趋势,2~4 h 内无显著差异,8~12 h 的金枕花粉活力高于猫山王,24 h 后基本丧失,表明常温干燥条件下花粉保存仅适用于 8 h 内的短期保存.亚历山大染色法更适合快速检测榴莲花粉活力以及活力高低,而离体培养萌发法虽耗时长但结果更准确可靠,二者结合可提高检测精确度.研究为优化人工榴莲授粉时间、制定花粉保存策略及推进杂交育种提供了科学依据.
In this study,two durian cultivars,'MonThong'(Jinzhen)and'Musang King'(Maoshanwang),were used as ex-perimental materials.Pollens at the full bloom stage were collected,and the pollen viability of durian stored for 2,4,8,12,18,and 24 hours was determined by the Alexander staining method and MTT staining method.In addition,the pollen germination rates at different culture times were observed via ex-vitro germination method(solid and liquid ex vitro germination methods).The results showed that the pollen viability of both cultivars showed a decreasing trend with the extension of storage time,with no significant difference within 2-4 hours;the pollen viability of'MonThong'(Jinzhen)was higher than that of'Musang King'(Maoshanwang)from 8 to 12 hours,and the pollen viability was basically lost after 24 hours,indicating that pollen storage under normal temperature and dry conditions is only suitable for short-term storage within 8 hours.The Alexander staining method is more suitable for the rapid detection of durian pollen viability and its level,while the ex-vitro germination method,although time-consuming,has more accurate and reliable results;the combination of the two methods can improve the detection accuracy.This study provides a scientific basis for optimizing the timing of artificial durian pollination,formulating pollen preservation strategies,and promoting hybrid breeding.
