珍稀植物蒜头果外植体消毒及诱导启动培养研究OA
Explant Disinfection and Induction Initiation Culture of the Rare Plants Malania oleifera
以蒜头果的顶芽和腋芽为实验材料,研究蒜头果外植体的无菌化消毒技术及诱导启动技术.结果表明,蒜头果外植体最佳的消毒方法为:75%酒精消毒1 min,无菌水清洗2次后,放在转速3 000~4 000 r/min的离心机上离心5 min,置于 NaClO 中消毒 5 min;此后用无菌水清洗 5 次,放到装有 0.1%HgCl2 的玻璃瓶中,消毒 10 min;最后用无菌水清洗 5 次.经过此消毒方法处理的外植体污染率低,有利于外植体转接及启动培养.最适合蒜头果诱导启动的培养基为改良 MS+6-BA 0.2 mg/L+NAA 0.2 mg/L,经此诱导出来的植株生长速度最快,分枝最多.
Taking the terminal buds and axillary buds of M.oleifera as experimental materials,aseptic disinfection technol-ogy and induction initiation culture technology for M.oleifera explants were studied.The results showed that the optimal dis-infection method for M.oleifera explants was as follows:disinfected with 75%alcohol for one min and rinsed twice with ster-ile water.the samples were centrifuged at a speed of 3 000~4 000 r·min-1 for five min,disinfected in NaClO for 5 min,and rinsed 5 times with sterile water.Then the samples were placed in a glass bottle containing 0.1%HgCl2,disinfected for 10 min,and finally rinsed five times with sterile water.The explants treated by this disinfection method had a low contamination rate,which was conducive to explant transfer and initiation culture.The most suitable medium for M.oleifera was induction initia-tion culture with modified MS+0.2 mg·L-1 6-BA+0.2 mg·L-1 NAA.The plants induced by this medium grew the fastest,had the most branches.
