EBV相关胃癌细胞中LMP2A对KLF5的表达调控作用OA

Objective To investigate the change in the expression of Krüppel-like factor 5(KLF5)in Epstein-Barr virus-as-sociated gastric carcinoma(EBVaGC)tissue and cells,as well as the regulatory effect of EBV-encoded latent membrane protein 2A(LMP2A)on KLF5 and the effect of KLF5 on the expression of the EBV lytic gene BZLF1 in EBVaGC cells.Methods Immu-nohistochemistry and Western blot were used to measure the expression of KLF5 in EBVaGC/EBV-negative gastric carcinoma(EBVnGC)tissue and cell lines.SGC7901 and SNU719 cells were transfected with LMP2A plasmid and siRNA,and the expression of KLF5 was observed.KLF5 was overexpressed in SNU719 cells,and the expression of BZLF1 was measured.Results The ex-pression level of KLF5 in EBVaGC tissue and cell lines was significantly lower than that in EBVnGC tissue and cell lines(t=6.125,P＜0.001).After LMP2A was overexpressed in SGC7901 cells,the expression of KLF5 was significantly downregulated(t=10.650,P＜0.001),and after LMP2A was knocked down in SNU719 cells,the expression of KLF5 was significantly upregulated(t=4.615,P＜0.01).Overexpression of KLF5 in SNU719 cells significantly upregulated the expression of BZLF1(t=5.741,P＜0.01).Conclusion EBV can inhibit the expression of KLF5 in EBVaGC tissue and cells.In EBVaGC cells,LMP2A can down-regulate the expression of KLF5,while KLF5 can upregulate the expression of the EBV lytic gene BZLF1.
