利塞膦酸钠对正畸大鼠牙根吸收及龈下菌群的影响OA
Effects of risedronate sodium on root resorption and subgingival microbiota in orthodontic rats
目的 探讨利塞膦酸钠(RS)对大鼠正畸诱导的炎性牙根吸收(OIIRR)及龈下菌群的影响及机制.方法 将50只Wistar大鼠随机分为正常对照组(Con组)、模型组、低剂量RS组、中剂量RS组和高剂量RS组,每组10只.除Con组外,其余各组均建立正畸牙移动模型,并给予相应剂量RS干预21 d.干预结束后,检测牙周临床指标;采用苏木精-伊红(HE)染色和抗酒石酸酸性磷酸酶(TRAP)染色观察组织形态并计数破骨细胞;采用酶联免疫吸附试验(ELISA)法检测牙周组织中炎症因子白细胞介素6(IL-6)、白细胞介素1β(IL-1β)和肿瘤坏死因子α(TNF-α)水平;结合细菌培养与 16S rRNA基因测序分析龈下菌群构成;采用Western blot方法检测牙周组织中核因子κB受体活化因子配体(RANKL)、骨保护素(OPG)和细胞外信号调节激酶(ERK)/丝裂原活化蛋白激酶(MAPK)信号通路关键蛋白表达.结果 与 Con组比较,模型组大鼠牙周组织出现细胞排列紊乱、炎性细胞浸润等病理改变,且其炎症因子水平、总细菌数、多种致病菌数量、多核破骨细胞数量、RANKL表达、RANKL/OPG比值、p-ERK和p-p38 MAPK蛋白表达水平均显著升高,而革兰阳性球菌数量降低,差异均有统计学意义(F=67.416~503.922,P<0.05).经RS干预后,上述异常指标均呈剂量依赖性改善(P<0.05).结论 RS可能通过抑制ERK/MAPK信号通路活化,减轻牙周炎症反应、调节龈下菌群失调并改善骨代谢失衡,从而缓解OIIRR.
Objective To investigate the effects and mechanisms of risedronate sodium(RS)on orthodontically induced in-flammatory root resorption and the subgingival microbiota in rats.Methods Fifty Wistar rats were randomly divided into a nor-mal control(Con)group,a model group,a low-dose RS group,a medium-dose RS group,and a high-dose RS group,with 10 rats in each group.Except for the Con group,orthodontic tooth movement models were established in all other groups,and correspon-ding doses of RS were administered for 21 days.After the intervention,periodontal clinical indicators were measured.Hematoxylin and eosin staining and tartrate-resistant acid phosphatase staining were used to observe tissue morphology and count osteoclasts.Enzyme-linked immunosorbent assay was employed to detect the levels of inflammatory cytokines interleukin-6,interleukin-1β,and tumor necrosis factor-α in periodontal tissues.Bacterial culture combined with 16S rRNA gene sequencing was utilized to ana-lyze the composition of the subgingival microbiota.Western blot was used to measure the expression of receptor activator of nuclear factor-KB ligand(RANKL),osteoprotegerin(OPG),and key proteins related to the extracellular signal-regulated kinase(ERK)/mitogen-activated protein kinase(MAPK)signaling pathway in periodontal tissues.Results Compared with the Con group,the periodontal tissues of rats in the model group showed pathological changes such as disorganized cell arrangement and inflammatory cell infiltration.Moreover,the levels of inflammatory cytokines,total bacterial count,counts of multiple pathogenic bacteria,number of multinucleated osteoclasts,RANKL expression,RANKL/OPG ratio,and protein expression levels of p-ERK and p-p38 MAPK were significantly increased in the model group,while the number of Gram-positive cocci significantly decreased(F=67.416-503.922,P<0.05).After RS intervention,the above abnormal indicators were improved in a dose-dependent manner(P<0.05).Conclusion RS may alleviate orthodontically induced inflammatory root resorption by inhibiting the activation of the ERK/MAPK signaling pathway,reducing periodontal inflammatory responses,regulating subgingival microbial dysbiosis,and a-meliorating bone metabolism imbalance.
