RcGSTF11作为潜在光合调节物的双效调节机制研究OA
Study on the Dual Regulatory Mechanism of RcGSTF11 Acting as a Potential Photosynthetic Regulator
为探明谷胱甘肽S-转移酶F亚家族(GSTF)作为潜在光合调节物参与光合作用的机制,在拟南芥中异源过表达蓖麻RcGSTF11基因.结果显示:RcGSTF11-OE拟南芥株高及角果产量均显著高于野生型,净光合速率(Pn)升高约28.31%;通过表面等离子体共振(SPR)与双分子荧光互补(BiFC)试验,验证了RcGSTF11与核酮糖-1,5-二磷酸羧化酶/加氧酶(Rubisco)大亚基RcRBCL之间存在相互作用,且RcGSTF11-OE拟南芥中Rubisco活性明显上升;转录组与代谢组学联合分析将差异基因富集在光合作用天线蛋白、光合作用及植物激素信号转导通路,相应地RcGSTF11-OE拟南芥中检测到糖类物质积累.此外,RcGSTF11的表达可以减少活性氧(ROS)的积累,提高还原型谷胱甘肽/氧化型谷胱甘肽(GSH/GSSG)比值,有助于维持植物体内氧化还原稳态并重塑下游基因的转录.综上所述,RcGSTF11被发现具有双重调节作用,近端与RBCL相互作用以调节光合碳同化,远端调节氧化还原稳态.
To investigate the mechanism through which GSTF acts as a potential photosynthetic regulator in photo-synthesis,this study overexpressed the castor bean RcGSTF11 gene in Arabidopsis thaliana.It was found that the plant height and silique yield of RcGSTF11-OE Arabidopsis plants were significantly higher than those of the wild type,and the net photosynthetic rate(Pn)increased by approximately 28.31%.The interaction between Rc GSTF11 and the large subunit of ribulose-1,5-bisphosphate carboxylase/oxygenase(Rubisco),Rc RBCL,was verified by surface plasmon reso-nance(SPR)and bimolecular fluorescence complementation(BiFC)assays.The Rubisco activity in RcGSTF11-OE Ara-bidopsis plants significantly increased.Combined analysis of transcriptomics and metabolomics enriched the differen-tially expressed genes in photosynthetic antenna proteins,photosynthesis,and plant hormone signal transduction path-ways.It was relatively found that sugar substances accumulated in RcGSTF11-OE Arabidopsis plants.In addition,the expression of RcGSTF11 can reduce the accumulation of ROS,increase the GSH/GSSG ratio,and help maintain the re-dox homeostasis in plants and reshape the transcription of downstream genes.In summary,Rc GSTF11 was found to have a dual regulatory effect of proximal interaction with RBCL to regulate photosynthetic carbon assimilation and distal regulation of redox homeostasis.
