3种传统发酵蔬菜制品中乳酸菌分离鉴定及其发酵特性分析OA
Isolation,Identification,and Characteristic Analysis of Lactic Acid Bacteria in Three Traditional Fermented Vegetable Products
为开发适用于云南酸笋发酵的纯种发酵剂,从3 种传统发酵蔬菜制品中分离筛选具有优良发酵性能的乳酸菌.采用 MRS 平板培养分离获得 3 株乳酸菌,分别命名为 LAB1、LAB2、LAB3;通过生理生化特性和 16S rDNA 测序鉴定菌种,并分析其发酵特性及功能特性.结果表明:LAB1、LAB3 为植物乳植杆菌(Lactiplantibacillus plantarum),LAB2 为哈尔滨乳杆菌(Lactobacillus harbinensis).菌株 LAB1 产酸能力较强,稳定期 pH≤3.60,且耐受 pH3.0 的环境,但 LAB3 在 pH3.0 的环境下生长受到抑制;菌株 LAB1、LAB3 耐盐性较强,能适应 8%盐浓度的环境;菌株 LAB2 的耐胆盐能力较强.3 株菌均具有较强的亚硝酸盐降解能力,培养 24 h 后降解率均超过 99%.3 株菌都有一定的抗氧化能力,对 DPPH 自由基和 ABTS+自由基的清除率分别为 9.13%~11.83%和 6.02%~8.37%;无细胞提取液的抗氧化能力较弱.3株菌均能有效抑制大肠杆菌和金黄色葡萄球菌,抑菌圈直径为 15.77~18.00 mm.以上结果表明,菌株 LAB1 和 LAB2 发酵性能优良,并具有降解亚硝酸盐和抑菌等特性,具有进一步制备酸笋纯种发酵剂的潜力.
To develop pure strain starter suitable for Yunnan sour bamboo shoot fermentation,lactic acid bacteria(LAB)with excellent fermentation properties are isolated and screened from three traditional fermented vegetable products.Three strains of LAB named LAB1,LAB2 and LAB3 are isolated by MRS plate culture,the strains are identified by physiological and biochemical characteristics and 16S rDNA sequencing,fermentation and functional properties are analysed.The results show that LAB1 and LAB3 are Lactobacillus plantarum,LAB2 is Lactobacillus harbinensis.Strain LAB1 has strong acid producing ability,pH≤3.60 in the stable period,and can tolerate the environment of pH3.0 but the growth of LAB3 is inhibited in an environment with pH of 3.0,strains LAB1 and LAB3 has a strong salt tolerance and could adapt to the environment of 8%salt concentration,strain LAB2 has strong bile salt tolerance.The results show that the three strains has strong nitrite degradation ability,and the degradation rate is more than 99%after 24 hours of culture.All three strains have some antioxidant capacity,and the scavenging rates of DPPH radicals and ABTS+radicals are 9.13%~11.83%and 6.02%~8.37%respectively,the antioxidant activity of acellular extract is weak.The three strains are able to inhibit Escherichia coli and Staphylococcus aureus,and the diameter of inhibition zone is from 15.77 to 18.00 mm.The above results show that the strains LAB1 and LAB2 have excellent fermentation performance,and have the characteristics of nitrite degradation and bacteriostasis,having the potential to further prepare pure strain starter of sour bamboo shoots.
