为挖掘优质微生物资源,从农家自制发酵辣椒中筛选分离发酵性能优良的乳酸菌,以期开发可应用于发酵蔬菜制品的纯种发酵剂.经平板分离培养得到 6 株乳酸菌,依次命名为 JX1-JX5、JS1,通过 16S rRNA 测序鉴定和菌株发酵特性、功能特性筛选评估,确定出候选发酵剂菌株.结果表明:JX1、JX5 为植物乳植杆菌(Lactiplantibacillus plantarum),JX2、JX3、JX4 为戊糖乳杆菌(Lactobacillus pentosus),JS1 为希腊魏斯氏菌(Weissella hellenica);JX1、JX3 产酸能力和耐酸能力最强,稳定期 pH≤3.80,耐受 pH 值为 3.0;菌株 JX3 耐盐性最强,10%盐浓度下 OD600值显著高于其他菌株;菌株 JX1-JX5 对亚硝酸盐的降解能力强,24 h 降解率均达到 96%以上.6 株菌都具有不同程度的体外抗氧化能力,其细胞悬浮液所具备的抗氧化能力均强于无细胞提取物;菌株 JX5 的细胞悬浮液清除 DPPH 能力最高,达到 14.67%;菌株 JS1 对羟自由基清除率最高,达到 55.74%;菌株 JX1 对超氧阴离子清除率最高,达到 51.17%;菌株 JX1、JX5 对大肠杆菌(E.coli)抑菌能力最强,菌株 JX3 对金黄色葡萄球菌(S.aureus)抑菌能力最强.本研究揭示出菌株 JX3 的优良发酵特性、很强的亚硝酸盐降解能力以及对 S.aureus一定的抑制能力,有望应用于后续纯种发酵剂的制备和开发.

To explore high-quality microbial resources,lactic acid bacteria with excellent fermentation performance are screened and isolated from homemade fermented chili peppers in order to develop purebred fermentation agents that can be applied to fermented vegetable products.Six strains of LAB are successfully isolated from the MRS solid culture medium through plate separation technique,designated as JX1-JX5 and JS1.These strains undergo identification via 16S rRNA sequencing,and comprehensive evaluation of their fermentation and functional characteristics.The sequencing results reveals that JX1 and JX5 are identified as Lactiplantibacillus plantarum,while JX2,JX3,and JX4 are classified as Lactobacillus pentosus.Additionally,JS1 is determined to be Weissella hellenica.Among these strains,both JX1 and JX3 exhibit the highest levels of acid production and during the stable period pH≤3.80,along with a pH tolerance of 3.0.Notably,strain JX3 demonstrate superior salt tolerance compared to other strains,displaying significantly higher OD600 values at a 10%salt concentration.The strains JX1-JX5 exhibit a high capacity for nitrite degradation,achieving a degradation rate of 96%within 24 h.The 6 strains exhibit varying degrees of in vitro antioxidant capacity,with the cell suspension demonstrating stronger antioxidant capacity compared to the cell-free extract.Strain JX5 display the highest DPPH clearance rate at 14.67%.Strain JS1 exhibit the highest hydroxyl free radical scavenging rate at 55.74%.The superoxide anion clearance rate is highest in strain JX1,reaching 51.17%.Both strains JX1 and JX5 demonstrate the strongest inhibitory capacity against E.coli,while strain JX3 show the strongest inhibitory capacity against S.aureus.In conclusion,strain JX3 display excellent fermentation characteristics,a strong ability to degrade nitrite,and certain inhibition capabilities against S.aureus,making it suitable for use in pure culture preparation.
