PCL/SBPU静电纺小口径人工血管的制备及性能研究OA
Preparation and performance research of PCL/SBPU electrospun small-diameter artificial blood vessel
为解决小口径人工血管与血液接触后引起血小板黏附等一系列问题,选用聚己内酯(PCL)为主要原料,通过静电纺丝技术制备PCL小口径人工血管,并将两性离子聚氨酯(SBPU)负载到PCL人工血管进行表面修饰,调控PCL与SBPU的共混比例,制备PCL、PCL/SBPU5、PCL/SBPU10及PCL/SBPU20共4种不同规格的人工血管.通过对其元素结构、微观形态、蛋白吸附能力等一系列的性能测试与表征,综合分析其抗血栓性能.结果表明:PCL/SBPU20的纤维微观形貌状态最佳.随着 SBPU含量增加,PCL/SBPU材料亲水性增加.PCL/SBPU20 的抗拉强度优于 PCL/SBPU5 和 PCL/SBPU10.PCL/SBPU5、PCL/SBPU10、PCL/SBPU20 这 3 组样品对 BSA 蛋白和 Fg 蛋白的吸附量均低于 PCL 样品.PCL/SBPU5、PCL/SBPU10、PCL/SBPU20这3组样品的L-LDH活力值均低于PCL样品.PCL、PCL/SBPU5、PCL/SBPU10、PCL/SBPU20这4组样品的相对细胞活性均大于80%.认为:SBPU表面修饰具有一定的抗蛋白吸附和抗血小板吸附效果,其中PCL/SBPU20的性能最佳.
To solve a series of problems such as platelet adhesion caused by the contact of small-diameter artificial blood vessels with blood,polycaprolactone(PCL)was selected as the main raw material.PCL small-diameter artificial blood vessels were prepared by electrospinning technology.Amphoteric ions polyurethane(SBPU)was loaded onto PCL artificial blood vessels for surface modification to regulate the blending ratio of PCL and SBPU.Four artificial blood vessels with different specifications were prepared,including PCL,PCL/SBPU5,PCL/SBPU10 and PCL/SBPU20.The antithrombotic performance was comprehensively analyzed through a series of performance tests and characterizations,such as its elemental structure,microscopic morphology and protein adsorption capacity.The results showed that the microstructure state of PCL/SBPU20 fibers was the best.The hydrophilicity of PCL/SBPU materials was increased as the increase of SBPU content.The tensile strength of PCL/SBPU20 was superior to that of PCL/SBPU5 and PCL/SBPU10.The adsorption capacities of BSA protein and Fg protein for the three groups of samples,namely PCL/SBPU5,PCL/SBPU10 and PCL/SBPU20,were all lower than those of the PCL sample.The L-LDH activity values of the three groups of samples,namely PCL/SBPU5,PCL/SBPU10 and PCL/SBPU20,were all lower than those of the PCL sample.The relative cell activities of the four groups of samples,namely PCL,PCL/SBPU5,PCL/SBPU10 and PCL/SBPU20,were all greater than 80%.It is believed that the surface modification of SBPU has certain anti-protein adsorption and anti-platelet adsorption effect,the performance of PCL/SBPU20 is the best.
