纤维素酶辅助提取皂荚皂苷的工艺优化及抗氧化活性研究OA
Optimization of Cellulase Assisted Extraction Process and Antioxidant Activity of Saponins From Gleditsia sinensis Lam.
通过优化皂荚皂苷的提取工艺,开发了一种提高皂苷提取得率的方法,并探究了皂荚皂苷的抗氧化能力.采用纤维素酶辅助提取皂荚皂苷,以皂荚皂苷的提取得率作为评价指标,在单因素试验基础上,通过响应面法优化了提取工艺条件,并测定了皂苷对DPPH自由基和ABTS自由基的清除能力,以探究其抗氧化能力.结果表明:加酶量3%(以荚果粉末质量计)、酶解温度50℃、酶解时间2.5 h、pH值5.0为皂荚皂苷提取的最佳工艺条件.在该条件下,皂荚皂苷的提取得率为26.457%,相较于未加纤维素酶的提取得率(11.01%)高出15.451个百分点.皂荚皂苷质量浓度在0.1~0.5 g/L范围内,对DPPH自由基有较好的清除作用;皂苷质量浓度在0.1~0.7 g/L范围内,对ABTS自由基有较好的清除作用,且清除率都随皂苷质量浓度的增加而升高.0.5 g/L皂苷溶液对DPPH自由基的清除率为80.81%,明显低于维生素C(Vc);而0.7 g/L皂苷溶液对ABTS自由基的清除率为96.63%,与Vc接近.纤维素酶辅助提取皂荚皂苷的提取得率高,且酶解法更绿色环保,最优工艺条件下得到的皂荚皂苷具有良好的抗氧化活性,因此该方法适合用于工业化生产.
By optimizing the extraction process of saponins from Gleditsia sinensis,a method was developed to improve the yield of saponin extraction and futher explored its antioxidant capacity.Using cellulase assisted extraction of G.sinensis saponins,the extraction yield of saponins was used as the evaluation index.Based on single factor experiments,the extraction process conditions were optimized by response surface methodology,and the scavenging ability of saponins on DPPH and ABTS free radicals was determined to explore their antioxidant capacity.The results showed that the optimal process conditions for extracting saponins from G.sinensis pods were 3%enzyme dosage(based on the mass of pod powder),enzymatic hydrolysis temperature of 50 ℃,enzymatic hydrolysis time of 2.5 h,and pH value of 5.0.Under these conditions,the extraction yield of saponins from G.sinensis was 26.457%,which was 15.451 percentage points higher than the extraction yield without cellulase(11.01%).The concentration of saponins in G.sinensis pods is within the range of 0.1-0.5 g/L,which has a good scavenging effect on DPPH free radicals.The concentration of saponins in the range of 0.1-0.7 g/L has a good scavenging effect on ABTS free radicals,and the scavenging rate increases with the increase of saponin concentration.The scavenging rate of 0.5 g/L saponin solution on DPPH free radicals is 80.81%,significantly lower than vitamin C(Vc),while the scavenging rate of 0.7 g/L saponin solution on ABTS free radicals is 96.63%,which is close to Vc.The extraction yield of saponins from G.sinensis assisted by cellulase is high,and the enzymatic hydrolysis method is more environmentally friendly.The saponins from G.sinensis obtained under the optimal process conditions have good antioxidant properties,and this method is suitable for industrial production.
