目的 探讨松乳菇多糖对乳腺癌细胞MDA-MB-231的增殖和凋亡的影响及分子机制.方法 将乳腺癌细胞株MDA-MB-231随机分为对照组,松乳菇多糖低、中、高剂量组,miR-NC组,miR-450b-3p组,松乳菇多糖+抑制剂阴性对照组,松乳菇多糖+miR-450b-3p抑制剂组;四甲基偶氮唑盐比色法检测细胞增殖抑制率;克隆形成实验检测细胞克隆形成能力;流式细胞术检测细胞凋亡;蛋白质印迹法检测蛋白表达;实时荧光定量聚合酶链反应检测miR-450b-3p表达水平.用松乳菇多糖和miR-450b-3p 抑制剂或抑制剂阴性对照处理乳腺癌异种移植小鼠,通过测量肿瘤体积、重量、miR-450b-3p表达及TUNEL阳性细胞率,以证实松乳菇多糖在体内的抗癌作用及机制.结果 松乳菇多糖低、中、高剂量处理的乳腺癌MDA-MB-231细胞克隆形成数下降,凋亡率、增殖抑制率、miR-450b-3p、胱天蛋白酶-3、胱天蛋白酶-9表达增高(P<0.05).miR-450b-3p过表达后,细胞克隆形成数下降,凋亡率、增殖抑制率、miR-450b-3p、胱天蛋白酶-3、胱天蛋白酶-9表达上升(P<0.05).抑制miR-450b-3p表达减弱了松乳菇多糖对MDA-MB-231细胞凋亡和增殖的影响.体内实验结果显示,松乳菇多糖降低肿瘤相对体积、肿瘤质量,提高miR-450b-3p表达及TUNEL阳性细胞率(P<0.05);沉默miR-450b-3p减弱了松乳菇多糖对移植瘤生长和凋亡的影响.结论 松乳菇多糖通过上调miR-450b-3p表达,抑制MDA-MB-231细胞增殖并诱导凋亡.

Objective To explore the effect of Lactarius deliciosus polysaccharide on the proliferation and apoptosis of breast cancer cell MDA-MB-231 and its molecular mechanism.Methods The breast cancer cell line MDA-MB-231 were randomly divided into a control group,low,medium,and high-dose Lactarius deli-ciosus polysaccharide groups,miR-NC group,miR-450b-3p group,and Lactarius deliciosus polysaccharide+anti-miR-NC group,Lactarius deliciosus polysaccharide+anti-miR-450b-3p group;the proliferation inhibition rate was detected by tetramethylazolium salt colorimetric method;clone formation experiment was used to detect cell clone formation ability;flow cytometry was used to detect apoptosis;Western blotting was used to detect protein expression;real-time fluorescent quantitative polymerase chain reaction was preformed to detect the expression of miR-450b-3p.Lactarius deliciosus polysaccharide and miR-450b-3p antagonist or antago-nist NC were used to treat xenografted mice with breast cancer.Tumor volume,mass,expression of miR-450b-3p and number of TUNEL positive cells were measured to confirm the anticancer effect and mechanism of Lactarius deliciosus polysaccharide in vivo.Results The number of clones formed in breast cancer MDA-MB-231 cells treated with low,medium,and high doses of Lactarius deliciosus polysaccharide was increased,and the apoptosis rate,proliferation inhibition rate,the expression levels of miR-450b-3p,cleaved-caspase-3 and cleaved-caspase-9 were increased(P<0.05).After an overexpression of miR-450b-3p,the number of clones formed was decreased,the apoptosis rate,proliferation inhibition rate,and the expression levels of miR-450b-3p,cleaved caspase-3 and cleaved caspase-9 were increased(P<0.05).Inhibition of miR-450b-3p expression weakened the effects of Lactarius deliciosus polysaccharide on the proliferation and apoptosis of breast cancer MDA-MB-231 cells.In vivo,Lactarius deliciosus polysaccharide decreased tumor relative volume and tumor weight,and increased the expression of miR-450b-3p and the rate of TUNEL positive cells(P<0.05).Silenc-ing miR-450b-3p reduced the effects of Lactarius deliciosus polysaccharide on the growth and apoptosis of transplanted tumors.Conclusion Lactarius deliciosus polysaccharide inhibits the proliferation of breast can-cer MDA-MB-231 and induces its apoptosis by up-regulating the expression of miR-450b-3p.
