马铃薯Y病毒SYRI株系外壳蛋白抗血清制备及应用OA
Preparation and application of antiserum against the coat protein of potato virus Y SYRI strain
[目的]马铃薯 Y病毒(potato virus Y,PVY)是烟草和马铃薯病毒病的主要病原,其 SYRI株系在中国属于优势株系,制备该株系外壳蛋白(coat protein,CP)抗血清可降低 PVY 的漏检率.[方法]以 PVY CPSYRI基因序列为模板进行扩增并连接原核表达载体 pEHISTEV,获得重组质粒 pEHISTEV-CPSYRI,将该质粒转化大肠杆菌 Rosetta,经原核表达后免疫新西兰白兔获得多克隆抗体.[结果]Western blot 结果表明制备的抗血清可检测到稀释 100 000倍的PVY CPSYRI蛋白,且不与同属的芜菁花叶病毒 CP蛋白发生反应.对田间马铃薯和烟草样品的检测表明,制备的抗血清能够有效地检测出田间发病植株.[结论]该抗血清具有良好的灵敏度和特异性,可应用于田间作物 PVY检测,为 PVY流行监测预警奠定了基础.
[Objective]Potato virus Y(PVY)is the main pathogen of tobacco and potato virus diseases.SYRI strain is the dominant strain in China.Preparation of coat protein(CP)antiserum of SYRI strain can reduce the missed detection rate of PVY.[Method]The PVY CPSYRI gene sequence was amplified and ligated into the prokaryotic expression vector pEHISTEV to obtain the recombinant plasmid pEHISTEV-CPSYRI.This recombinant plasmid was then transformed into Escherichia coli Rosetta,and after prokaryotic expression,New Zealand white rabbits were immunized to obtain polyclonal antibodies.[Result]Western blot results showed that the prepared antiserum could detect PVY CPSYRI protein diluted 100 000-fold,and it exhibited no cross-reaction with the CP protein of Turnip mosaic virus(TuMV),which belonged to the same genus.Tests on potato and tobacco samples in the field showed that the antiserum prepared was effective in detecting the diseased plants in the field.[Conclusion]The antiserum has showed good sensitivity and specificity,and could be used to detect PVY in crops in the field,which has laid a foundation for the monitoring and early warning of PVY epidemic.
