基于GSK-3β/β-catenin通路探究健脾散结方对结肠癌大鼠肿瘤生长及炎症反应的影响OA
Exploration on effects of Jianpi Sanjie prescription on tumor growth and inflammatory response in rats with colon cancer based on GSK-3β/β-catenin pathways
为探究健脾散结方对结肠癌大鼠肿瘤生长及炎症反应的影响及机制,用1,2-二甲肼诱导结肠癌大鼠模型,将48只Wistar大鼠随机分为对照组、模型组、健脾散结方低、中、高剂量组、健脾散结方+氯化锂组,每组8只.记录各组大鼠体质量变化,测量结肠长度及统计肿瘤数量.采用HE染色观察结肠组织病理变化,免疫组化法检测结肠组织增殖细胞核抗原(PCNA)表达,酶联免疫吸附法检测血清炎症因子含量,Western blotting法检测结肠组织糖原合成酶激酶-3β/β-连环蛋白(GSK-3β/β-catenin)通路蛋白表达.结果表明:与对照组比较,模型组大鼠体质量、结肠长度、血清IL-10、结肠组织p-GSK-3β/GSK-3β水平显著减少,肿瘤数、PCNA阳性表达细胞数、血清白细胞介素(IL-6、IL-1β)、肿瘤坏死因子α(TNF-α)、β-catenin表达显著增加(P<0.05);与模型组比较,健脾散结方中、高剂量组体质量、结肠长度、血清IL-10、结肠组织p-GSK-3β/GSK-3β水平显著增加,肿瘤数、PCNA阳性表达细胞数、IL-6、IL-1β和TNF-α、β-catenin表达显著减少(P<0.05);与健脾散结方高剂量组比较,健脾散结方+氯化锂组体质量、结肠长度、血清IL-10、结肠组织p-GSK-3β/GSK-3β水平显著减少,肿瘤数、PCNA阳性表达细胞数、血清IL-6、IL-1β和TNF-α、β-catenin表达显著增加(P<0.05).综上,健脾散结方可能通过激活GSK-3β/β-catenin信号通路,减轻炎症反应,从而抑制结肠癌大鼠肿瘤生长.
In order to explore the effects and mechanism of Jianpi Sanjie prescription on tumor growth and inflammato-ry response in rats with colon cancer,modes of colon cancer rat were induced by 1,2-dimethylhydrazine,and 48 Wistar rats were randomly divided into control group,model group,low-dose,medium-dose and high-dose Jianpi Sanjie prescrip-tion groups,and Jianpi Sanjie prescription+lithium chloride group,8 cases in each group.The changes of body mass were recorded,colon length and number of tumors were measured.The pathological changes of colon tissues were observed by HE staining,expression of proliferating cell nuclear antigen(PCNA)in colon tissues was detected by immunohisto-chemistry,levels of serum inflammatory factors were detected by enzyme-linked immunosorbent assay,and expressions of glycogen synthase kinase-3β/β-catenin(GSK-3β/β-catenin)pathway related proteins in colon tissues were detected by Western blotting.The results showed that compared with control group,body mass,colon length,levels of serum IL-10 and p-GSK-3β/GSK-3β in colon tissues were significantly decreased,number of tumors and PCNA-positive cells,levels of serum interleukin(IL-6,IL-1β)and tumor necrosis factor α(TNF-α),and β-catenin expression were significantly in-creased in model group(P<0.05).Compared with model group,body mass,colon length levels of serum IL-10 and p-GSK-3β/GSK-3β in colon tissues were significantly increased,number of tumors and PCNA-positive cells,levels of IL-6,IL-1β and TNF-α,and β-catenin expression were significantly decreased in medium-dose and high-dose Jianpi Sanjie prescription groups(P<0.05).Compared with high-dose Jianpi Sanjie prescription group,body mass,colon length,levels of serumIL-10 and p-GSK-3β/GSK-3β in colon tissues were significantly decreased,number of tumors and PCNA-positive cells,levels of IL-6,IL-1β and TNF-α,and β-catenin expression were significantly increased in Jianpi Sanjie prescription+lithi-um chloride group(P<0.05).In conclusion,Jianpi Sanjie prescription may relieve inflammatory response by activating GSK-3β/β-catenin signaling pathways,and thus inhibit tumor growth in rats with colon cancer.
