硫酸葡聚糖抑制ALV-J复制及其机制初探OA
Inhibitory effect of dextran sulfate on ALV-J replication and preliminary exploration of its mechanism
为评估硫酸葡聚糖(DS)对J亚群禽白血病病毒(ALV-J)复制的抑制效果及其潜在机制,采用体内、外试验相结合的方法进行系统研究.通过灌胃给予健康雏鸡低(0.2 g·kg-1)、中(1 g·kg-1)、高(5 g·kg-1)剂量的DS进行安全性评价,观察其对雏鸡生长状态和体重的影响.建立ALV-J感染雏鸡模型,以相同剂量的DS进行干预,采用qPCR检测肝脏、脾脏、法氏囊等免疫器官中的病毒载量.体外试验中,采用MTT和CCK-8法评估DS对鸡巨噬细胞系HD11的细胞毒性,并通过qPCR和Western blot试验检测DS对ALV-J感染HD11细胞后病毒包膜蛋白(env)基因转录与蛋白质表达的影响,同时分析炎症因子IL-1β的表达水平.结果表明:DS在0.2~5 g·kg-1剂量范围内对HD11细胞无明显的细胞毒性,雏鸡灌胃后亦未出现明显的毒副作用,生长状态正常;DS干预可显著降低ALV-J感染雏鸡免疫器官中的病毒载量,并显著下调HD11细胞中ALV-J env基因mRNA和蛋白质的表达水平;此外,DS能以浓度和时间依赖性的方式上调IL-1β的表达.综上,DS可通过激活宿主炎症反应(上调IL-1β)有效抑制ALV-J的复制,且具有良好的安全性,为开发防控禽白血病的新型免疫调节剂提供理论依据.
In order to evaluate the inhibitory effect of dextran sulfate(DS)on the replication of avian leukosis virus subgroup J(ALV-J)and its potential mechanism,a systematic study was conducted by combining in vitro and in vivo ex-periments.The safety of low(0.2 g·kg-1),medium(1 g·kg-1)and high(5 g·kg-1)doses of DS was evaluated by gavage to healthy chicks,and the effects on the growth status and weight of chicks were observed.Subsequently,a chick-en model infected with ALV-J was established,and the same dose of DS was used to intervene.The viral load in immune organs such as liver,spleen and bursa of Fabricius was detected by qPCR.In vitro,MTT and CCK-8 methods were used to evaluate the cytotoxicity of DS to chicken macrophage cell line HD11,qPCR and Western blot were used to detect the effects of DS on the gene transcription and protein expression of virus envelope protein(env)after ALV-J infected HD11 cells,and the expression level of inflammatory factor IL-1β was also analyzed.The results showed that DS had no obvious cytotoxicity to HD11 cells in the dose range of 0.2-5 g·kg-1,and the chicks had no obvious toxic and side effects after gavage,and their growth status was normal.DS intervention can significantly reduce the viral load in immune organs of chickens infected with ALV-J,and significantly reduce the mRNA and protein expression level of ALV-J env gene in HD11 cells.In addition,DS can up-regulate the expression of IL-1β in a concentration and time-dependent manner.To sum up,DS can effectively inhibit the replication of ALV-J by activating the host inflammatory response(up-regulating IL-1β),and it has good safety,which provides a theoretical basis for the development of new immunomodulators for the prevention and treatment of avian leukemia.
