低温处理对不同栽培区蓝莓枝条生理特性与花芽形态的影响OA

为探明不同栽培区蓝莓品种的休眠特性差异,并优化区域化栽培管理技术.以吉林、山东、湖北、云南4个栽培区的7个蓝莓品种为试验材料,通过低温处理结合人工培养,对花芽萌发进程的影响进行观测,并利用石蜡切片技术观察其花芽形态特征和组织分化过程.结果表明:随低温处理时间的延长,枝条中可溶性糖含量呈逐渐上升趋势,淀粉含量整体降低,可溶性蛋白呈现先下降后上升的趋势,游离氨基酸含量呈先上升后下降的趋势.抗氧化酶活性在休眠解除中期达到峰值,丙二醛含量呈先缓慢上升后逐渐降低趋势.人工培养条件下蓝莓横纵径变化结果表明:不同栽培区蓝莓品种在人工培养条件下花芽横纵径均呈现显著增长趋势,但品种间和地区间存在显著差异.蓝莓品种在花芽分化初期与末期的形态变化,通过石蜡切片观察结果显示:在分化初期花芽内部结构紧密,细胞分裂活动较弱,花器官原基不明显,分化后期可以清晰观察到花萼、花瓣、柱头和雄蕊的原基结构,细胞分裂活动达到高峰.不同品种花器官分化的时间和程度存在差异,与其低温处理和光照培养时长有关.该结果可为不同气候区域的蓝莓品种选择、精准控温栽培技术优化以及新品种抗寒性评价提供理论依据和技术参考.

In order to explore the differences of dormancy characteristics of blueberry varieties in dif-ferent cultivation areas and optimize the regional cultivation and management technology,seven blue-berry varieties in four cultivation areas of Jilin,Shandong,Hubei and Yunnan were used as test mate-rials in this study.The effects of low-temperature treatment combined with artificial culture on the germination process of flower buds were dynamically observed,and the morphological characteristics and tissue differentiation process of flower buds were observed by paraffin section technique.The re-sults showed that the soluble sugar content in the branches showed a gradual upward trend,the starch content decreased as a whole,the soluble protein showed a first decreasing and then increas-ing trend,and the free amino acid content showed a first increasing and then decreasing trend.The activity of antioxidant enzymes reached the peak at the middle stage of dormancy release,and the content of malondialdehyde increased slowly first and then decreased gradually.The results of dy-namic changes of horizontal and vertical diameters of blueberry under artificial culture conditions showed that the horizontal and vertical diameters of flower buds of blueberry varieties in different cultivation areas showed a significant growth trend under artificial culture conditions,but there were significant differences among varieties and regions.The morphological changes of blueberry variet-ies at the early and late stages of flower bud differentiation were observed by paraffin section.The re-sults showed that the internal structure of flower buds was close,the cell division activity was weak,and the primordium of flower organs was not obvious at the early stage of differentiation.At the later stage of differentiation,the primordium structure of calyx,petal,stigma and stamen could be clearly observed,and the cell division activity reached the peak.The time and degree of floral organ differ-entiation of different varieties were different,which was related to the duration of low temperature treatment and light culture.The results of this experiment can provide theoretical basis and techni-cal reference for the selection of blueberry varieties,the optimization of precise temperature control cultivation techniques and the evaluation of cold resistance of new varieties in different climatic re-gions.
