Comparative Study on Gut Bacteria Diversity of Captive and Wild Rana dybowskii in Liaoning Province

为研究圈养与野生环境下东北林蛙肠道细菌群落结构和功能的差异,采集辽宁地区野生和圈养东北林蛙各12只,分别取其十二指肠、回肠和直肠组织,采用16S rDNA高通量测序技术鉴定菌群结构,通过alpha多样性分析、beta多样性分析和聚类分析方法比较2组间菌群差异.结果表明:野生东北林蛙肠道优势菌门为变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)和放线菌门(Actinobacteria),优势菌属为分枝杆菌属(Mycobacterium)、假单胞菌属(Pseudomonas)和拟杆菌属(Bacteroides);圈养组优势菌门为变形菌门、厚壁菌门和拟杆菌门(Bacteroidetes),优势菌属为假单胞菌属、柠檬酸杆菌属(Citrobacter)和希瓦氏菌属(Shewanella).在东北林蛙全肠道中,变形菌门、放线菌门和拟杆菌门为优势菌门,野生组变形菌门的相对丰度低于圈养组.野生与圈养东北林蛙整体肠道细菌alpha多样性差异显著(P<0.05),beta多样性差异较小;对野生和圈养东北林蛙肠道不同部位的细菌多样性进行比较,回肠部位细菌的alpha多样性和直肠部位的beta多样性差异较大.研究结果为探究不同环境对东北林蛙肠道健康状态以及功能的影响提供依据.

To investigate the differences in the structure and function of gut bacterial communities between captive and wild Rana dybowskii,12 wild Rana dybowskii and 12 captive Rana dybowskii were collected from Liaoning Province.Samples were taken separately from the duodenum,ileum,and rectum.The microbial community structure was identified using high-throughput sequencing technology for 16S rDNA.Alpha diversity analysis,beta diversity analysis,and cluster analysis were used to compare the differences in microbial communities between the two groups.The results showed that the dominant phyla in the intestinal tract of wild Rana dybowskii were Proteobacteria,Firmicutes,and Actinobacteria,while the dominant genera were Mycobacterium,Pseudomonas,and Bacteroides.The dominant phyla in the captive group were Proteobacteria,Firmicutes,and Bacteroidetes,while the dominant genera were Pseudomonas,Citrobacter,and Shewanella.In the entire gut of Rana dybowskii,the phyla Proteobacteria,Actinobacteria,and Bacteroidetes were the dominant phyla,the relative abundance of Proteobacteria in the wild group was lower than that in the captive group.There was a significant difference in the overall gut bacterial alpha diversity between wild and captive groups of Rana dybowskii(P<0.05),while the beta diversity difference was small.Comparing the bacterial diversity of different intestinal parts between wild and captive Rana dybowskii,the alpha diversity of bacteria in the small intestine and the beta diversity in the rectum differed obviously.The research results provide a scientific basis for investigating the effects of different environments on the intestinal health status and gut function of the Rana dybowskii.
