Optimization of Fermentation Conditions for Clostridium butyricum HADIG-CB004 by Response Surface Methodology

为提高丁酸梭菌 HADIG-CB004 的生物量,开发一种经济高效的培养基,研究结合单因素试验与响应面分析,对其发酵培养基组成进行优化.单因素试验结果表明,葡萄糖、酵母浸粉、鱼蛋白胨和柠檬酸钠可显著促进菌体生长;响应面分析表明,丁酸梭菌 HADIG-CB004 的最佳培养条件为葡萄糖 13.77 g/L、酵母浸粉 8.99 g/L、鱼蛋白胨 11.05 g/L、柠檬酸钠 10.00 g/L、硫酸铵 1.00 g/L、L-半胱氨酸盐酸盐 1.00 g/L、一水硫酸锰 0.25 g/L、初始pH值 6.8.该条件下菌液OD600 值达 3.98,活菌数为5×108 CFU/mL,分别较传统强化梭菌培养基提高67.9%和28.2%.优化后的丁酸梭菌增殖培养基成分简单、成本较低,适用于丁酸梭菌的营养体扩繁及后生元制备.

To develop a cost-effective medium for enhancing the biomass of Clostridium butyricum HADIG-CB004,this study employed single-factor experiments and response surface methodology(RSM)to optimize the medium composition.Single-factor experiments results showed that glucose,sodium citrate,yeast extract,and fish peptone could significantly promote bacterial growth.RSM results showed that the optimal medium was composed of 13.77 g/L glucose,8.99 g/L yeast extract,11.05 g/L fish peptone,and 10.00 g/L sodium citrate,1.00 g/L ammonium sulfate,1.00 g/L L-cysteine hydrochloride,and 0.25 g/L manganese sulfate monohydrate,with initial pH value of 6.8.Validation experiments showed that under these conditions,the OD600 value reached 3.98,and the viable bacterial count reached 5×108 CFU/mL,which increased by 67.9%and 28.2%,respectively,compared with those under culture with the conventional reinforced Clostridium medium.The optimized C.butyricum proliferation medium with simple components and a low cost was suitable for the propagation of C.butyricum and preparation of postbiotics.
