红景天及其活性成分防治神经退行性病变药理作用的研究进展OA
Research progress on the pharmacological effects of Rhodiola and its active components in preventing and treating neurodegenerative diseases
中药红景天在防治神经退行性病变方面具有多途径、多靶点的特点.红景天及其活性成分可通过降低β-淀粉样蛋白的沉积,减少Tau蛋白过度磷酸化,阻止α-突触核蛋白聚集,激活细胞自噬,减轻神经毒性.红景天及其活性成分可激活DJ-1/核因子E2相关因子2、核因子E2相关因子2/血红素加氧酶-1信号通路,提高NADPH氧化酶2的活性,减轻氧化应激反应.红景天及其活性成分可抑制核因子-κB信号通路活化,阻止核苷酸结合寡聚化结构域样受体蛋白3炎症小体形成,下调多种促炎因子的分泌,调节肠—脑轴,减轻炎症反应.红景天及其活性成分可调控凋亡相关蛋白,激活磷脂酰肌醇3-激酶/蛋白激酶B、核因子E2相关因子2/谷胱甘肽过氧化物酶4信号通路,抑制神经元凋亡.红景天及其活性成分可上调脑源性神经营养因子的表达,抑制丝裂原活化蛋白激酶信号通路过度活化,增强神经可塑性.红景天及其活性成分可改善胆碱能神经功能,促进学习记忆功能恢复.红景天及其活性成分可促进三磷酸腺苷的合成,促进线粒体自噬,改善线粒体功能,对神经功能发挥保护作用.本文整理了近年来红景天活性成分用于神经退行性病变的相关报道,总结其药理作用,以供临床及后续研究参考.
As a traditional Chinese medicine,Rhodiola is characterised by multi-pathway and multi-target in preventing and treating neurodegenerative diseases.Rhodiola and its active components can reduce the deposition of β-amyloid,decrease tau protein hyperphosphorylation,prevent α-synuclein aggregation,activate autophagy,and alleviate neurotoxicity.Rhodiola and its active components can enhance the activity of NADPH oxidase 2 and alleviate oxidative stress by activating the DJ-1/nuclear factor E2-related factor 2(NF-E2-related factor 2)/heme oxygenase-1 signaling pathways.Rhodiola and its active components can inhibit the activation of the nuclear factor-kappa B(NF-κB)signaling pathway,prevent the formation of the NOD-like receptor protein 3(NLRP3)inflammasome,reduce the secretion of various pro-inflammatory factors,regulate the gut-brain axis,and alleviate inflammatory responses.Rhodiola and its active components can regulate apoptosis-related proteins,activate the phosphatidylinositol 3-kinase(PI3K)/Akt and Nrf2/glutathione peroxidase 4(GPX4)signaling pathways,and inhibit neuronal apoptosis.Rhodiola and its active components can upregulate the expression of brain-derived neurotrophic factor,inhibit the excessive activation of the mitogen-activated protein kinase(MAPK)signaling pathway,and enhance neural plasticity.Rhodiola and its active components can improve cholinergic neuronal function and promote the recovery of learning and memory functions.Rhodiola and its active components can promote the synthesis of adenosine triphosphate,enhance mitophagy,improve mitochondrial function,and exert a protective effect on neural function.This review summarizes recent reports on the application of Rhodiola and its active components in the prevention and treatment of neuro-degenerative diseases,and summarizes their pharmacological effects to provide a reference for clinical practice and future research reference.
