基于DNA重测序筛选加系西门塔尔牛超数排卵效果相关候选基因研究OA
Study on Candidate Genes Related to the Effect of Ovulation Induced by DNA Resequencing Screening in Simmental Cattle
超数排卵是牛胚胎生产的重要环节,但供体牛对超数排卵处理的反应存在显著个体差异,其遗传基础尚未完全阐明.本研究以60头16月龄的健康加系西门塔尔供体牛为研究对象,在饲养管理条件和超数排卵程序相同的前提下,根据胚胎数量和胚胎可用率构建高、低表型群体,采集血液样品开展全基因组DNA重测序.通过群体固定系数(FST)和遗传多样性比值(Pi ratio)联合进行选择消除分析,系统筛选与超数排卵效果相关的候选基因.结果表明,共鉴定出10个可能参与或调控超数排卵过程的候选基因,其中BMPR1B、MMP 1、PGR、GHR、CD36和ERBB4与排卵数量显著相关,SOX5、CAMK1D、HDAC4和BCL2与胚胎可用率显著相关.上述基因主要涉及卵泡发育、颗粒细胞增殖及卵母细胞成熟等生物过程.研究结果为解析西门塔尔牛超数排卵性状的遗传基础及供体牛精准筛选提供了分子依据.
Superovulation is an important step in the production of bovine embryos,but there are significant individual differences in the responses of donor cattle to superovulation treatment,and its genetic basis has not been fully clarified.This study took 60 16-month-old healthy Canadian Simmental donor cattle as the research subjects.Under the conditions of unified feeding management and superovulation program,high and low phenotypic populations were constructed based on the number of embryos and embryo availability.Blood samples were collected for whole-genome DNA resequencing.Through the combined selection elimi-nation analysis of population fixation index(FST)and genetic diversity ratio(Pi ratio),the candidate genes related to the effect of superovulation were systematically screened.The results indicated that a total of 10 candidate genes that might be involved in or regulate the process of superovulation were identified.Among them,BMPR1B,MMP 1,PGR,GHR,CD36 and ERBB4 were significantly correlated with the number of ovulation,while SOX5,CAMK1D,HDAC4 and BCL2 were significantly correlated with the availability of embryos.The above-mentioned genes are mainly involved in biological processes such as follicular deve-lopment,granulosa cell proliferation and oocyte maturation.The research results provide a molecular basis for analyzing the genetic basis of the superovulation trait in Simmental cattle and for precise screening of donor cattle.
