目的:探讨壮骨胶囊(ZGC)调控Ras蛋白(RAS)/Raf蛋白激酶(RAF)丝裂原激活蛋白激酶(MEK)/细胞外信号调节激酶(ERK)通路对骨质疏松症(OP)大鼠骨代谢的影响.方法:将60只大鼠随机分为正常组、模型组、壮骨胶囊低剂量(ZGC-L)组(0.5 g/kg)、壮骨胶囊高剂量(ZGC-H)组(2.0 g/kg)、ZGC-H(2.0 g/kg)+ML-099(11.7 mg/kg)组,每组12只.给药12周后测量各组大鼠股骨的骨密度(BMD);HE染色检测股骨组织病理变化;TUNEL染色检测股骨细胞凋亡;酶联免疫吸附试验(ELISA)检测血清中骨特异性碱性磷酸酶(BALP)、骨钙素(OC)、核心结合因子-α1(CBF-α1)、Ⅰ型原胶原N端前肽(PINP)水平,蛋白质印迹法(Western blotting)检测RAS/RAF/MEK/ERK信号通路相关蛋白的表达.结果:正常组大鼠骨小梁排列整齐、致密,间隙均匀,骨组织结构完整且形态规整;模型组大鼠骨小梁间隙较大,股骨组织稀疏,排列紊乱,股骨组织损伤明显;ZGC-L组、ZGC-H组大鼠骨小梁间距较窄,股骨组织相对紧密,形态结构规整,股骨组织损伤降低;ZGC-H+ML-099组大鼠股骨组织损伤程度高于ZGC-H组.模型组大鼠股骨细胞凋亡率、股骨RAS、RAF蛋白相对表达量及p-MEK1/2/MEK1/2、p-ERK1/2/ERK1/2高于正常组(P<0.05),股骨BMD及血清BALP、OC、CBF-α1、PINP水平低于正常组(P<0.05);ZGC-L组、ZGC-H组大鼠股骨细胞凋亡率、股骨RAS、RAF蛋白相对表达量及p-MEK1/2/MEK1/2、p-ERK1/2/ERK1/2低于模型组(P<0.05),股骨BMD及血清BALP、OC、CBF-α1、PINP水平高于模型组(P<0.05);ZGC-H+ML-099组大鼠股骨细胞凋亡率、股骨RAS、RAF蛋白相对表达量及p-MEK1/2/MEK1/2、p-ERK1/2/ERK1/2高于ZGC-H组,股骨BMD及血清BALP、OC、CBF-α1、PINP水平低于ZGC-H组(P<0.05).结论:壮骨胶囊可能通过抑制RAS/RAF/MEK/ERK通路调节OP大鼠骨代谢.

Objective:To investigate the effect of Zhuanggu capsule(ZGC)on bone metabolism in rats with osteoporosis(OP)by regulating the rat sarcoma(RAS)/Raf protein kinase(RAF)/mitogen activated protein kinase(MEK)/extracellular regulated protein kinase(ERK)pathway signaling pathway.Methods:A total of 60 rats were randomly divided into a normal group,a model group,a low-dose ZGC(ZGC-L)group(0.5 g/kg),a high-dose ZGC(ZGC-H)group(2.0 g/kg),and a ZGC-H(2.0 g/kg)+ML-099(11.7 mg/kg)group,with 12 rats in each group.After 12 weeks of administration,the bone mineral density(BMD)of the femur was measured in each group.Pathological changes of femoral tissue were detected by HE staining.Femoral cell apoptosis was detected by TUNEL staining.The serum levels of bone-specific alkaline phosphatase(BALP),osteocalcin(OC),core-binding factor-α1(CBF-α1),and procollagen type I N-terminal propeptide(PINP)were measured by enzyme linked immunosorbent assay(ELISA).The expression of proteins related to the RAS/RAF/MEK/ERK signaling pathway was detected by Western blotting.Results:In the normal group,the trabecular bone was arranged neatly and densely with uniform gaps,and the bone tissue structure was intact and regular.In the model group,the trabecular bone gaps were larger,the femoral tissue was sparse and disorderly arranged,and the femoral tissue damage was obvious.In the ZGC-L group and ZGC-H group,the trabecular bone spacing was narrower,the femoral tissue was relatively dense and structurally regular,and the femoral tissue damage was reduced.The degree of femoral tissue damage in the ZGC-H+ML-099 group was higher than that in the ZGC-H group.Compared with the normal group,the model group showed a higher femoral cell apoptosis rate,higher relative expression levels of femoral RAS and RAF proteins,higher p-MEK1/2/MEK1/2 and p-ERK1/2/ERK1/2 ratios(P<0.05),lower femoral BMD,and lower serum levels of BALP,OC,CBF-α1,and PINP(P<0.05).Compared with the model group,the ZGC-L group and ZGC-H group showed a lower femoral cell apoptosis rate,lower relative expression levels of femoral RAS and RAF proteins,lower p-MEK1/2/MEK1/2 and p-ERK1/2/ERK1/2 ratios(P<0.05),higher femoral BMD,and higher serum levels of BALP,OC,CBF-α1,and PINP(P<0.05).Compared with the ZGC-H group,the ZGC-H+ML-099 group showed a higher femoral cell apoptosis rate,higher relative expression levels of femoral RAS and RAF proteins,higher p-MEK1/2/MEK1/2 and p-ERK1/2/ERK1/2 ratios(P<0.05),lower femoral BMD,and lower serum levels of BALP,OC,CBF-α1,and PINP(P<0.05).Conclusion:Zhuanggu capsule may regulate bone metabolism in OP rats by inhibiting the RAS/RAF/MEK/ERK signaling pathway.
