ABF(ABRE binding factors)/AREB(ABA responsive element binding protein)转录因子在植物生长发育和非生物胁迫响应中发挥重要作用.为初步探究芜菁ABF基因在其肉质根发育过程中的作用,本研究采用生物信息学方法对芜菁ABF/AREB基因家族成员、蛋白的理化性质、基因结构和保守基序、染色体定位、启动子顺式作用元件、蛋白互作网络等进行分析;并分别利用转录组数据和实时荧光定量PCR(qRT-PCR)技术分析BcABF基因在芜菁肉质根不同发育时期的表达特征和表达水平.结果表明,共鉴定到15个ABF/AREB家族成员,可划分为3个亚家族(Group A、Group B和Group C),随机分布在8条染色体上.顺式作用元件分析结果表明BcABF包含大量生长发育、非生物胁迫与激素应答元件,转录组分析发现肉质根膨大过程中Group A家族成员基因表达水平显著高于Group B和Group C.通过qRT-PCR分析芜菁不同发育时期肉质根中6个BcABF基因的表达水平,发现BcABF2基因在次生根增厚期(RTS)显著高表达,推测BcABF2可能是影响芜菁肉质根膨大的关键基因.本研究为深入挖掘调控肉质根膨大的核心BcABF基因功能与芜菁品种选育奠定了理论依据.

ABF(ABRE binding factors)/AREB(ABA responsive element binding protein)transcription factors play crucial roles in plant growth,development,and abiotic stress responses.This study aimed to preliminarily investigate the functions of ABF genes during fleshy root development in turnip(Brassica rapa).Bioinformatics approaches were employed to identify and characterize members of ABF/AREB gene family in turnip,including analysis of protein physicochemical properties,gene structure and conserved motifs,chromosomal localization,promoter cis-acting elements,protein-protein interaction networks.Transcriptome data and quantitative real-time PCR(qRT-PCR)were used to analyze the expression patterns and levels of BcABF genes across different developmental stages of turnip fleshy roots,respectively.A total of 15 ABF/AREB family members were identified and classified into three subfamilies:Group A,Group B and Group C.These members were randomly distributed across eight chromosomes.Cis-acting element analysis revealed that BcABF promoters possess numerous elements related to growth and development,abiotic stress,and hormone response.Transcriptome analysis indicated that the expression levels of Group A members were significantly higher than those of Group B and C during fleshy root expansion.qRT-PCR analysis of six BcABF genes at different developmental stages of the fleshy roots of turnip revealed that BcABF2 gene was highly expressed specifically during the secondary root thickening stage(RTS),suggesting it may be a key regulator of turnip fleshy root expansion.These results provide a foundation for further functional characterization of key BcABF genes involved in promoting turnip fleshy root expansion and for variety improvement through molecular breeding.
