木槿SSR标记开发及杂种后代鉴定OA
Development of Hibiscus syriacus SSR Markers and Identification of Hybrids
[目的]为木槿育种及品种鉴定提供技术支撑,本研究基于木槿转录组数据开发简单序列重复(SSR)分子标记,并将其用于杂交后代的分子鉴定.[方法]以不同品种木槿的花瓣为试验材料,利用 Illumina 平台进行转录组测序,结合 MISA 在线工具分析 SSR 位点,采用 Primer-BLAST 在线软件设计特异性引物.通过试剂盒法提取木槿叶片总脱氧核糖核酸(DNA),以12 组亲本 DNA 为模板进行聚合酶链反应(PCR)扩增,经琼脂糖凝胶电泳检测,筛选出多态性引物.[结果]本研究共获得17 098 个 SSR 位点,涵盖99 种重复基元,各类型基元种类数差异明显,其中单核苷酸、三核苷酸和六核苷酸重复基元种类数分别为2、10 和38.筛选出60 个序列长度≥30 bp 的 SSR 位点,并从中获得3 个多态性良好的 SSR 标记(HBG18、HBG7 和 HBG21),成功鉴定了 5 个杂交组合('玫瑰木槿'×'中国雪纺'、'蓝鸟'×'汉帛'、'粉红心重瓣'×'中国雪纺'、'普粉'×'红心'、'蓝色雪纺'×'长苞重瓣')F1 代的真实性.[结论]本研究开发的SSR 标记具有较高的多态性和分辨率,为木槿分子标记辅助选择提供了候选分子标记,有助于推动木槿属植物的分子标记辅助育种及种质资源保护.
[Objective]To support Hibiscus syriacus breeding and variety identification,this study de-veloped SSR markers based on transcriptome data and applied them for the molecular identification of hy-brid offspring.[Method]In this study,petals from different H.syriacus varieties were used as experi-mental materials.Transcriptome sequencing was performed using the Illumina platform,followed by anal-ysis of SSR loci using the MISA online tool.Specific primers were designed with Primer-BLAST online software.Total DNA was extracted from H.syriacus leaves using a kit method.PCR amplification was conducted using genomic DNA from 12 parent plants as templates,and the products were detected via agarose gel electrophoresis.Finally,polymorphic primers were screened.[Result]A total of 17 098 SSR sites were obtained,exhibiting diverse repeat motifs comprising 99 types.The number of motif types var-ied considerably among different categories:there were 2 types of mononucleotides,10 types of trinucle-otides,and 38 types of hexanucleotides.Sixty SSR loci with sequence lengths of 30 bp or more were se-lected.Three SSR markers with high polymorphism(HBG18,HBG7,and HBG21)were further devel-oped and successfully used to verify the authenticity of F1 progeny from five hybrid combinations:'Meiguimujin'×'Zhongguoxuefang','Lanniao'×'Hanbo','Fenhongxinchongban'×'Zhongguoxue-fang','Pufen'×'Hongxin','Lansexuefang'×'Changbaochongban'.[Conclusion]The SSR markers developed in this study exhibit high polymorphism and resolution,providing candidate markers for marker-assisted selection and promoting molecular-assisted breeding and germplasm conservation of Hibis-cus species.
