白酒酒曲中淀粉酶产生菌筛选与产酶条件研究OA

酒曲中微生物种类繁多,酶系资源丰富,显著影响酒的品质.以市售白酒酒曲为原料,以透明圈直径和淀粉酶活力为评价指标,从酒曲中分离并筛选优良的产淀粉酶菌株.系统探究了发酵时间、发酵温度、发酵液pH值、培养基碳氮比和接种量等因素对菌株产酶的影响,通过正交实验优化产酶条件.结果表明,筛选所得菌株的最适产酶条件为:发酵时间96 h,发酵温度30℃,发酵液pH值7.0,培养基碳氮比2∶5,菌液接种量为4%.在此条件下,淀粉酶酶活达到90.6 U·mL-1.

Chinese fermentation starter harbors diverse microbial communities and rich enzyme systems that significantly influence liquor quality.Taking transparent circle diameter and amylase activity as evaluation indicators,this study isolated and screened superior amylase-producing strains from commercially available Chinese fermentation starter The effects of fermentation time,fermentation temperature,pH,carbon-nitrogen(C/N)ratio,and inoculation amount on results were systematically investigated,orthogonal experiments were conducted to optimize the enzyme production conditions.The results showed that the optimal conditions were:fermentation time of 96 h,fermentation temperature of 30℃,pH of 7.0,C/N ratio of 2∶5,and inoculum volume of 4%,and the amylase activity reached 90.6 U·mL-1.This study provides a theoretical basis for microbial screening from Chinese fermentation starter and fermentation production of related metabolites.
