为探究绿原酸(chlorogenic acid,CGA)在湖羊生发泡(GV)期卵母细胞玻璃化冷冻保存和解冻后体外成熟过程中的连续保护作用,试验将湖羊GV期卵母细胞分为新鲜对照组、冷冻对照组、低浓度CGA(50 μmol/L)组和高浓度CGA(100 μmol/L)组.分别检测解冻后卵母细胞存活率、体外成熟后第一极体(PB1)排出率、活性氧(ROS)与谷胱甘肽(GSH)水平、超氧化物歧化酶(SOD)与过氧化氢酶(CAT)活性、线粒体膜电位(MMP)及ATP水平,并评估体外受精后的卵裂率与囊胚率.结果显示:与新鲜对照组相比,冷冻对照组卵母细胞存活率、PB1排出率、GSH相对水平、抗氧化酶SOD与CAT相对活性、MMP相对水平、ATP相对水平、卵裂率及囊胚率均极显著降低(P<0.01或P<0.001),ROS相对水平极显著升高(P<0.001).与冷冻对照组相比,低浓度CGA组卵母细胞存活率、PB1排出率、GSH相对水平、抗氧化酶SOD与CAT相对活性、MMP相对水平、ATP相对水平、卵裂率及囊胚率显著(P<0.05)或极显著升高(P<0.01或P<0.001),ROS相对水平显著降低(P<0.05).然而高浓度CGA组上述指标与冷冻对照组相比均无显著差异(P>0.05).综上所述,在湖羊卵母细胞玻璃化冷冻保存和解冻后体外成熟过程中添加50 μmol/L CGA可通过抑制氧化应激、维持抗氧化酶活性及改善线粒体功能,提高湖羊GV期卵母细胞冷冻解冻后存活、成熟及后续发育能力.

To investigate the continuous protective effects of chlorogenic acid(CGA)during vitrification cryopreservation and post-thawing in vitro maturation(IVM)of Hu sheep germinal vesicle(GV)stage oocytes,GV oocytes were collected and divided into four groups:fresh control,frozen control,frozen-thawed low-concentration CGA(50 μmol/L),and frozen-thawed high-concentration CGA(100 μmol/L).Survival rate after thawing,first polar body(PB1)extrusion rate after IVM,reactive oxygen species(ROS)and glutathione(GSH)levels,superoxide dismutase(SOD)and catalase(CAT)activities,mitochondrial membrane potential(MMP),and ATP levels were measured,and cleavage and blastocyst rates following in vitro fertilization(IVF)were evaluated.The results showed that the oocyte survival rate,PB1 extrusion rate,relative GSH level,relative activities of antioxidant enzymes SOD and CAT,relative MMP level,relative ATP level,cleavage rate,and blastocyst rate were significantly lowered(P<0.01 or P<0.001)compared with the fresh control group,whereas the relative ROS level was significantly elevated(P<0.001).Compared with the frozen control group,the low-concentration CGA group showed significant improvements in oocyte survival rate,PB1 extrusion rate,relative GSH levels,relative activities of antioxidant enzymes SOD and CAT,relative MMP levels,relative ATP levels,cleavage rate,and blastocyst rate(P<0.05,P<0.01 or P<0.001),and a significant decrease in relative ROS levels(P<0.05).In contrast,no significant differences were detected in any parameters mentioned above between the high-concentration CGA group and the frozen control group(P>0.05).In conclusion,supplementation with 50 μmol/L CGA during vitrification cryopreservation and subsequent IVM of Hu sheep oocytes enhances frozen-thawed survival,maturation,and subsequent developmental competence by inhibiting oxidative stress,maintaining antioxidant enzyme activities,and improving mitochondrial function.
