益气活血化痰方对慢性阻塞性肺疾病大鼠肺功能及炎症因子的影响OA
Effects of Yiqi Huoxue Huatan Formula on Lung Function and Inflammatory Factors in Rats with Chronic Obstructive Pulmonary Disease
目的:探讨益气活血化痰方(Yiqi Huoxue Huatan Formula,YHHF)对慢性阻塞性肺疾病(chronic obstructive pulmonary dis-ease,COPD)大鼠肺功能及炎症因子的改善作用.方法:将46 只 SD 大鼠随机分为对照组11 只和造模组35 只,后者采用香烟烟雾熏吸联合气管内滴注脂多糖法制备 COPD 模型.造模成功后,大鼠随机分为模型组、YHHF 低剂量组(YHHF-L 组,14.94 g·kg-1)、YHHF 中剂量组(YHHF-M 组,29.88 g·kg-1)及 YHHF 高剂量组(YHHF-H 组,59.76 g·kg-1),每组 8只.灌胃30 d 后,检测大鼠肺功能指标,包括呼吸频率(respiratory frequency,RF)、潮气量(tidal volume,TV)、每分钟气量(mi-nute volume,MV)、吸气峰值(peak inspiratory flow,PIF)、呼气峰值(peak expiratory flow,PEF)及呼气中段流速(expiratory flow at 50%,EF50).采用 ELISA 法检测大鼠血清白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-6(interleukin-6,IL-6)及肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)含量.采集大鼠支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)并进行细胞分类计数;采用 HE 染色和 Masson 染色检测大鼠肺组织病理形态.结果:与对照组比较,模型组大鼠肺功能指标TV、PIF、PEF、MV、EF50 均降低,RF 升高(P<0.05),血清 IL-1β、IL-6、TNF-α 含量及 BALF 中细胞总数、嗜酸性粒细胞、中性粒细胞、淋巴细胞、巨噬细胞计数均升高(P<0.05),肺组织呈明显炎症病理改变.与模型组比较,YHHF-H 组大鼠 TV、PIF、PEF、MV、EF50 均升高,RF 降低(P<0.05),血清 IL-1β、IL-6 含量及 BALF 中细胞总数、嗜酸性粒细胞、中性粒细胞、淋巴细胞、巨噬细胞计数均降低(P<0.05),肺组织病理损伤减轻,纤维化程度改善.结论:YHHF 可改善 COPD 大鼠肺功能,降低炎症因子水平,减轻肺组织病理损伤.
Objective:To investigate the effects of Yiqi Huoxue Huatan Formula(YHHF)on lung function and inflammatory factors in rats with chronic obstructive pulmonary disease(COPD).Methods:Forty-six SD rats were randomly divided into a control group(11 rats)and a modeling group(35 rats),the latter of which was used to establish a COPD model through cigarette smoke exposure com-bined with intratracheal instillation of lipopolysaccharide.After model establishment,the rats were randomly divided into a model group,a low-dose YHHF group(YHHF-L,14.94 g·kg-1),a medium-dose YHHF group(YHHF-M,29.88 g·kg-1),and a high-dose YHHF group(YHHF-H,59.76 g·kg-1),with 8 rats in each group.After 30 days of gavage,lung function indicators of the rats were measured,including respiratory frequency(RF),tidal volume(TV),minute volume(MV),peak inspiratory flow(PIF),peak ex-piratory flow(PEF),and expiratory flow at 50%(EF50).ELISA was used to detect the levels of serum interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)in rats.Bronchoalveolar lavage fluid(BALF)was collected for cell classification and counting.Hematoxylin and eosin(HE)staining and Masson staining were used to observe the pathological morphology of rat lung tissue.Results:Compared with the control group,the model group rats showed decreased lung function parameters including TV,PIF,PEF,MV,and EF50,and increased RF(P<0.05).Serum IL-1β,IL-6,TNF-α levels,as well as total cell count,eosino-phils,neutrophils,lymphocytes,and macrophage counts in BALF were all elevated(P<0.05),and lung tissue showed obvious inflam-matory pathological changes.Compared with the model group,rats in the YHHF-H group showed increased TV,PIF,PEF,MV,EF50,and decreased RF(P<0.05);serum IL-1β and IL-6 levels,and total cell count,eosinophils,neutrophils,lymphocytes,and macro-phage counts in BALF were decreased(P<0.05);lung tissue pathological damage was alleviated,and fibrosis was improved.Conclu-sion:YHHF can improve lung function in COPD rats,reduce levels of inflammatory factors,and alleviate pathological damage in lung tissue.
